The World Health Organization's global initiative toward eliminating high-risk Human Papillomavirus (hrHPV)-related cancers recommends DNA testing over visual inspection in all settings for primary cancer screening and HPV eradication by 2100. However, multiple hrHPV types cause different of cancers, there is a pressing need an easy-to-use, multiplex point-of-care diagnostic platform detecting types. Recently, CRISPR-Cas systems have been repurposed detection. Here, we established multiplexed assay (CRISPRD) to detect cervical cancer-causing hrHPVs one reaction (one-pot assay). We harnessed the compatibility thermostable AapCas12b, TccCas13a, HheCas13a nucleases with isothermal amplification successfully detected HPV16 HPV18, along internal control single-pot limit detection 10 copies 100% specificity. This offers rapid practical solution hrHPVs, which may facilitate large-scale screening. Furthermore, CRISPRD programmability enables it be adapted any two nucleic acid biomarkers as well control.

Human papillomavirus (HPV) infection is a global public health concern and associated with cervical cancer. HPV genotype mapping has an essential role in prevention control strategy developing more suitable vaccine for Indonesia. This was descriptive retrospective cross-sectional study from 2012 until 2022 at Kalgen Laboratory, Jakarta all over the metropolitan regions. The total 76,413 samples were collected consecutive sampling, which 694 excluded, thus final used 75,719. DNA test performed using polymerase chain reaction (PCR): SPF10-DEIA-LiPA25 methods. genotyping procedures included extraction, PCR XpressMatrix kit, hybridization. From 75,719 samples, 93.4% negative intraepithelial lesion or malignancy (NILM). Among 6.6% of abnormal cytology groups, 53.8% atypical squamous cells undetermined significance (ASCUS), 32.9% low grade (LSIL), 13.3% high (HSIL). most common risk genotypes among HSIL 16, 18, 52, 58, 33, 51, 53. Single compared to multiple infections. showed that HR-HPV types 52 frequent type NILM, ASCUS, LSIL. Thus, it could serve as potential future reference create nonavalent Indonesian population based on its different epidemiology.

LCMV has been engineered development of cancer immunotherapies, currently undergoing evaluation phase I/II clinical trials. Initial findings have demonstrated safety an exceptional ability activate expand tumor-specific T lymphocytes. Combination strategies maximize antitumor effectiveness LCMV-based immunotherapies are being explored. Methods We assessed therapeutic effects intratumoral administration polyinosinic:polycytidylic acid (poly(I:C)) systemic vaccination using LCMV-vector expressing non-oncogenic versions E6 E7 antigens human papillomavirus 16 (artLCMV-E7E6) a bilateral model engrafting TC-1/A9 cells. This cell line, derived from parental TC-1, exhibits low MHC class I expression is highly immune-resistant. The mechanisms underlying combination’s efficacy were investigated through bulk RNA-seq, flow cytometry analyses tumor microenvironment, selective depletions antibodies clodronate liposomes, Batf3 deficient mice, vivo bioluminescence experiments.
