Abstract Chemotherapy, as a conventional strategy for tumor therapy, often leads to unsatisfied therapeutic effect due the multi-drug resistance and serious side effects. Herein, we genetically engineered thermal-responsive murine Ferritin (mHFn) specifically deliver mitoxantrone (MTO, chemotherapeutic photothermal agent) tissue chemotherapy combined therapy of colorectal cancer, thanks high affinity mHFn transferrin receptor that highly expressed on cells. The thermal-sensitive channels allowed effective encapsulation MTO in vitro laser-controlled release vivo. Upon irradiation with 660 nm laser, raised temperature triggered opening channel nanocage, resulting controlled rapid MTO. Consequently, significant amount reactive oxygen species was generated, causing mitochondrial collapse cell death. photothermal-sensitive release, low systemic cytotoxicity, excellent synergistic eradication ability vivo made mHFn@MTO promising candidate chemo-photothermal combination against cancer. ConspectusThe significance of cancer stem cells (CSCs), a rare population in tumor tissues, biology and the treatment solid malignancies has been widely appreciated for more than two decades. Due to peculiar self-renewal capability, even one single cell can grow into bulk mass. For this reason, CSCs have long blamed as major culprit initiation, progression, resistance, metastasis, recurrence. Therefore, it postulated that targeting could provide tremendous clinical benefits patients with tumors. Accumulating studies corroborated maintained tight regulation redox homeostasis fate was extremely sensitive elevated oxidative stress. Accordingly, plethora therapeutic drugs generate reactive oxygen species (ROS) leveraged target CSCs.
