Abstract Taxanes are first-line chemotherapeutic agents for patients with triple-negative breast cancer (TNBC). However, resistance, the main cause of clinical treatment failure and poor prognosis, reduces their effectiveness has become an increasingly important problem. Recently, a form iron-dependent programmed cell death called ferroptosis was reported to play role in regulating tumor biological behavior. In this study, we revealed prognostic significance ferroptosis‑related gene (FERG) model clarified that ferroptosis-related genes may be promising candidate biomarkers therapy. First, resistance-related FERGs were screened, univariate Cox regression analysis used construct model, including GRIK3, IDO1, CLGN. Then, TNBC TCGA database classified into high-risk low-risk groups. The identification high scores had higher probability dying earlier than those low scores. Moreover, these three associated immune infiltrates checkpoints patients. conclusion, study suggested significantly chemotherapy resistance can as predictors possibly targeted therapy future.

Breast cancer arises from the malignant transformation of mammary epithelial cells under influence various carcinogenic factors, leading to a gradual increase in its prevalence. This disease has become cause mortality among female malignancies, posing significant threat health women. The timely identification breast remains challenging, often resulting diagnosis at advanced stages disease. Conventional therapeutic approaches, such as surgical excision, chemotherapy and radiotherapy, exhibit limited efficacy controlling progression metastasis Regulated cell death (RCD), process essential for physiological tissue renewal, occurs within body independently external influences. In context cancer, research on RCD primarily focuses cuproptosis, ferroptosis pyroptosis. Mounting evidence suggests marked association between these specific forms RCD, onset cancer. For example, cuproptosis vector can effectively bind copper ions induce cells, thereby hindering their proliferation. Additionally, expression ferroptosis‑related genes enhance sensitivity chemotherapy. Likewise, pyroptosis‑related proteins not only participate pyroptosis, but also regulate tumor microenvironment, ultimately cells. present review discusses unique regulatory mechanisms pyroptosis through which they are affected by conventional drugs. Furthermore, it provides comprehensive overview significance modulating highlights shared characteristics. knowledge may provide novel avenues both clinical interventions fundamental

The buildup of lipid peroxides on the cell membrane is critical in initiation Ferroptosis, an iron-dependent form controlled death. Ferroptosis a type death that varies from other types both mechanics and morphology, it holds significant promise for cancer therapy. As result, there has been increasing interest research community regarding exploration understanding recent years. This review article aims to provide solid theoretical foundation management cancer. It accomplishes this by summarizing processes contribute development outlining underlying mechanisms various tumors.

Background: Most patients with advanced-stage breast cancer present bone metastasis, which seriously affects their quality of life and prognosis. We aimed to investigate the potential role mechanism circular Forkhead box protein P1 (FOXP1) (hsa_circ_0008234) in metastasis cancer. Methods: The Gene Expression Omnibus database (GEO) (GSE111504) was used screen differentially expressed RNAs (circRNAs) metastatic related expression FOXP1 (circFOXP1) miRNA measured by real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR). Cellular experiments were performed assess influence circFOXP1 cells. After co-culture siRNA-transfected MDA-MB-231 cells marrow-derived mesenchymal stem (BMSCs), effect on osteogenic genes detected. Bioinformatic Ontology Kyoto Encyclopedia Genes Genomes enrichment analyses using clusterProfilter 4.8.2 R package version 4.3. Results: upregulated cancer, particularly Silencing decreased abilities proliferation, migration, invasion. increased alkaline phosphatase (ALP) activity gene BMSCs co-cultured MDA-MB-231/si-circRNA group observed. miR-338-3p a target circFOXP1. analysis indicated that targeted mRNAs involved MAPK pathway, regulation actin cytoskeleton, tight junction, Ras PI3K-AKT pathway. Conclusions: upregulation might repress cellular activities facilitate osteogenetic differentiation microenvironment targeting miR-338-3p. be therapeutic for
