Many studies on osteoblasts have suggested that Wnt5a plays a crucial role in excessive osteoblast activity, which is responsible for ectopic new bone formation, but research osteoclasts ankylosing spondylitis (AS) remains relatively limited. This study aimed to explore whether influences osteoclastmediated resorption curdlan-injected SKG mice, model mimics AS.Compared the Vehicle group, treatment group exhibited statistically higher clinical arthritis scores and increased hindpaw thickness values.Microcomputed tomography (microCT) analysis of hindpaws revealed significant increase inflamed density Wnt5a-treated compared group.Histological examination also showed pronounced inflammation structural damage marrow ankles group.Intriguingly, microCT femur trabecular loss was markedly observed group. Both number TRAP-positive their activity were greater group.Serum markers resorption, not significantly elevated group.Notably, promotion osteoclast differentiation by inhibited following with anti-Wnt5a.These findings suggest targeting could be promising strategy mitigating pathological features AS modulating activity.

Ginsenosides, bioactive compounds from the genus Panax, have potential therapeutic effects on diverse ailments, including diabetes. Emerging evidence suggests their involvement in bone metabolism. The present review summarizes current understanding of ginsenosides osteoporosis, periodontal disease, and osteoarthritis. Their mechanisms action include osteoblasts, osteoclasts, ligament fibroblasts (PDLFs), chondrocytes, which are pivotal maintaining bone, tissue, cartilage homeostasis. Ginsenosides may exert beneficial by enhancing PDLF osteoblast activity, suppressing osteoclast function, augmenting chondrocyte synthesis matrix, mitigating connective tissue degradation. Moreover, they possess antioxidant, anti-inflammatory, antimicrobial, anti-pyroptotic properties. efficacy increasing density, ameliorating periodontitis, alleviating osteoarthritis symptoms has been demonstrated preclinical studies using animal models. In terms mechanism action, modulate cellular differentiation, key signaling pathway molecules, such as mitogen-activated protein kinases (MAPKs), while also regulating various mediators. Furthermore, symptomatic relief observed models lends further credence to utility. However, translate these findings into clinical practice, rigorous investigations imperative ascertain safety, efficacy, optimal dosing regimens human subjects.

Zishen Yuzhen Pill (ZYP) is a Chinese herbal product developed by Shenzhen TCM Hospital, which have been frequently used to treat osteoporosis (OP). This study aimed determine the major chemical components of ZYP and its prototype compounds metabolites in rat biological samples, as well explore potential effect ZYP-containing serum MC3T3-E1 cells. UPLC-Q/TOF-MS was identify components. Then, orally administered rat, samples plasma, urine, feces, bile, tissue were collected metabolites. The viability cells evaluated using CCK-8 method after treatment with various concentrations (2%, 4%, 8%) serum. Following serum, activity alkaline phosphatase (ALP) Alizarin red S (ARS) measured, levels Runx2, Opn, Opg Osterix quantified qPCR Western blot analysis. And for RNA-seq Results indicated that total 152 identified ZYP, including flavonoids, iridoid, lignans, triterpene saponins, etc. Furthermore, we detected 70 99 distributed different tissues. In addition, observably promoted osteogenesis increasing ALP ARS activities, up-regulating expression results beneficial effects may be related upregulation mitochondrial oxidative phosphorylation. work provided further support traditional application OP. this can promote pharmacokinetic pharmacological evaluation ZYP.

Osteoporosis (OP) is a metabolic bone disease with high incidence rate worldwide. Its main features are decreased mass, increased fragility and deterioration of microstructure. It caused by an imbalance between formation resorption. Ginsenoside safe effective traditional Chinese medicine (TCM) usually extracted from ginseng plants, having various therapeutic effects, which the effect against osteoporosis has been extensively studied. We searched total 44 relevant articles using keywords including osteoporosis, ginsenosides, mesenchymal cells, osteoblasts, osteoclasts remodeling, all investigated cellular mechanisms different types ginsenosides affecting activity remodeling stem osteoblasts to counteract osteoporosis. This review describes used treat perspectives, providing solid theoretical basis for future clinical applications.

Background Fu-zi decoction (FZD) has a long history of application for treating Rheumatoid arthritis (RA) as classic formulation. However, its underlying mechanisms have not been fully elucidated. This study aimed to decipher the potential mechanism FZD in RA, with specific focus on receptor activator nuclear factor κB/receptor κB ligand (RANK/RANKL) signaling pathway. Methods The impact RA was investigated collagen-induced rats (CIA), and an osteoclast differentiation cell model. In vivo , antiarthritic effect at various doses (2.3, 4.6, 9.2 g/kg/day) evaluated by index score, paw volume, toe thickness histopathological examination inflamed joints. Additionally, ankle joint tissues were determined micro-CT safranin O fast green staining evaluate synovial hyperplasia articular cartilage damage. vitro maturation TRAP RANKL-induced bone marrow mononuclear cells. levels pro- anti-inflammatory cytokines well RANKL OPG ELISA kits. addition, Western blotting used investigate RANK/RANKL pathway activation both . Results significantly diminished weigh loss CIA rats, alleviated pathological alterations. Consistent results, markedly inhibited decreasing numbers dose-dependent manner. Moreover, decreased pro-inflammatory IL-6, IL-1β TNF-α, while increasing cytokine IL-10 level serum culture supernatants. Treatment reduced levels, increased RANKL/OPG ratio. settings, downregulated protein expressions RANK, RANKL, c-Fos, elevating further Conclusion conclusion, exerts therapeutic inhibiting RANK/RANKL-mediated differentiation, which suggested that is promising treatment RA.
