Finding efficient therapeutic strategies to fight antibiotic-resistant bacteria is a complicated affair specifically in the therapy of chronic bacterial infections related hospital-acquired infections. Recently, three major antibacterial systems based on antisense RNA, CRISPR-Cas9, and metal/metal oxide nanoparticles particularly silver (Ag) have shown more effective activity compared conventional antibiotics. Abstract Nanotechnology has paved the way for novel treatment strategies diseases such as cancer. While chemical synthesis of nanoparticles (NPs) can introduce toxic and expensive reducing reagents, green offers an alternative with dual capping properties. Curcumin exhibits favourable pharmacological properties, enabling synergism therapeutic cargo. This study aimed to compare chemically synthesised gold NPs delivery F Luc -mRNA in vitro . Chemical AuNPs were produced using trisodium citrate curcumin, respectively, functionalised poly-L-lysine (PLL) polyethylene glycol (PEG). The NP:mRNA nanocomplexes characterised nanoparticle tracking analysis (NTA), transmission electron microscopy (TEM), Fourier transform infrared (FTIR), UV–visible spectroscopy. Various mRNA binding studies assessed optimal binding, compaction, protection mRNA. All small (<150 nm) had good colloidal stability, protection. MTT (3-[4,5-dimethylthiazol-2-yl]-2.5-diphenyltetrazolium bromide) assay showed cell viability, significant transgene expression noted luciferase reporter gene assay. Higher transfection was achieved human cervical carcinoma (HeLa) than breast adenocarcinoma (MCF-7) embryonic kidney (HEK293) cells. Both curcumin-synthesised displayed similar activity all cells, curcumin-capped marginally better at same concentration.
