Perceptual awareness of auditory stimuli decreases from wake-fulness to sleep, largely due reduced cortical responsiveness. During wakefulness, neural responses external exhibit a broader spatiotemporal propagation pattern compared deep sleep. A potential mechanism for this phenomenon is the synaptic upscaling excitatory connections during as posited by homeostasis hypothesis. However, we argue that uniform alone cannot fully account observation. We propose novel suggesting between different areas exceeds within individual wakefulness. Our computational results demonstrate former promotes transfer and information, whereas latter has diminishing effects. Neuronal interactions give rise to complex dynamics in cortical networks, often described terms of the diversity activity patterns observed a neural signal. Interestingly, complexity spontaneous electroencephalographic signals decreases during slow-wave sleep (SWS); however, underlying mechanisms remain elusive. Here, we analyse in-vivo recordings from neocortical and hippocampal neuronal populations rats show that decrease is due emergence synchronous DOWN states. Namely, find states SWS force population be more recurrent, deterministic, less random than REM or wakefulness, which, turn, leads field recordings. Importantly, when exclude analysis, wakefulness become indistinguishable: spiking all collapses common scaling. We complement these results by implementing critical branching model cortex, which shows inducing only percentage neurons enough generate replicates SWS.

Abstract Preclinical behavioral testing is essential for drug discovery in neuropsychiatric disorders, yet translational challenges persist because of interspecies differences. Touchscreen‐based tasks offer a promising solution bridging this gap. These provide flexibility across cognitive domains and species, facilitating rigorous comparisons. They complement traditional assays, offering improved face, predictive, construct validity by mirroring human neuropsychological tests. Notably, nearly identical have been validated multiple enhancing potential. Recent studies demonstrate conserved neurocircuitry engagement touchscreen tasks, supporting their relevance to function therapeutic development. The integration electrophysiological measures, such as electroencephalography (EEG) local field potential (LFP) recordings with enhances biomarker serves elucidate neural circuit dynamics. Despite current limitations the need further validation, approach offers pathway more efficient discovery. This review covers recent research describing feasibility benefits EEG/LFP‐touchscreen combination rodents. While still its early stages, promise strategy evident. Future efforts will likely focus on refining methodologies, identifying robust biomarkers, expanding species. platforms, integrated measurements, hold significant advance our understanding disorders accelerate development effective treatments. image
