Introduction: Microbes of sponges have diverse associations, including true symbiosis. Sponges, being evolutionarily ancient sessile filter feeders, host and abundant microbial species that play crucial roles in metabolism. Although the symbionts are widely distributed within organism (up to 40% their volume), ecological relationships interactions between bacteria sponge remain largely unexplored for many species. The present study was one first attempts isolate symbiotic from Raspaciona aculeata. Materials Methods: After isolation on marine agar medium, isolates were characterized different colony morphology. 16S rDNA taxonomic analysis carried out isolates. Results: Following an incubation period two weeks at 25°C, only 13 bacterial strains isolated with a very low rate genetic biodiversity. All belonged Gammaproteobacteria class (Pseudomonadaceae family), except (isolate AL-18ra) belonging Bacilli (Bacillaceae family). Conclusion: obtained results great importance advancing understanding symbiosis phenomena aculeata its bioapplication potential.

Abstract The capacity of marine sponges to cope with environmental changes is often attributed the specific composition their bacterial communities. In this study, we assessed microbiome two dominant sponges, Rhabdastrella globostellata (Rg) and Hyrtios erectus (He), living in Bouraké lagoon (New Caledonia), where abiotic conditions daily fluctuate according tide. Sponge-associated bacteria have been mostly cultured from shallow water (≤30 m) sponges, whereas only few studies targeted specimens below 30 m. This study assessed the cultivability of two marine sponges Xestospongia muta and Agelas sventres collected (<30 m), upper mesophotic (30–60 lower (60–90 reefs. were cultivated on six different media, replicate plates used to pick individual colonies or recover entire biomass. Prokaryotic community analysis was conducted using Illumina MiSeq sequencing 16S rRNA gene amplicons. In this study, we used 16S rRNA gene amplicon sequencing to investigate prokaryotic community composition of the Caribbean sponges Xestospongia muta and Agelas sventres from three depth ranges: < 30 m (shallow), 30-60 (upper mesophotic), 60-90 (lower mesophotic). The in shallow samples X. was enriched Cyanobacteria, Chloroflexota, Crenarchaeota compared mesophotic depths, while were Acidobacteriota. For A. sventres, relative abundance Acidobacteriota, Gammaproteobacteria higher samples, Proteobacteria samples. Antimicrobial activity evaluated by screening crude extracts against a set Gram-positive Gram-negative bacteria, yeast, an oomycete. Recent reviews have reinforced sponge-associated bacteria as a valuable source of structurally diverse secondary metabolites with potent biological properties, which makes these microbial communities promising sources new drug candidates. However, the overall diversity metabolite biosynthetic potential present in is difficult to access due fact that majority are not readily cultured laboratory. Thus, use cultivation-independent approaches may allow accessing “silent” and “cryptic” gene clusters cannot yet be cultured. In study, we investigated (BGCs) metagenomes bacterial associated three sponge species: Clathria reinwardti, Rhabdastrella globostellata, Spheciospongia sp. The results reveal contain high number predicted BGCs, ranging from 282 463 BGCs per metagenome. types were represented 12 different cluster types. Clusters encode fatty acid synthases polyketide (PKS) most dominant BGC types, followed by encoding synthesis terpenes bacteriocins. Based on sequence similarity analysis, 363 families (GCFs) identified. Species misidentification in the field of natural products is an acknowledged problem. These errors are especially widespread sponge studies, albeit rarely assessed and documented. As a case study, we aim to revisit reports isomalabaricane triterpenes, isolated from four demosponge genera: Jaspis, Geodia, Stelletta Rhabdastrella. From total 44 articles (1981-2022), 27 unique vouchers were listed, 21 which accessed re-examined here: 11 (52.4%) these misidentified. Overall, 65.9% studies published incorrect species name: previously identified Jaspis all fact Rhabdastrella globostellata. We conclude that triterpenes only two possibly one Geodia species. This work aims to characterize the microbial diversity of encrusting sponge Cliona varians, a pore-forming and coral reef bioeroding marine emerging spread related ocean acidification.We analysed microbiome composition by 16S V4 amplicon next-generation sequencing in community encrusting/excavating varians thriving at Southern Caribbean Sea.
