Abstract Fluorescence imaging in the second near‐infrared window (NIR‐II, 1000–1700 nm) has aroused immense attention for biomedical applications, offering exceptional advantages such as ultra‐low photon scattering and increased tissue penetration. Among NIR‐II‐emitted organic dyes, Boron dipyrromethene (BODIPY), emerged a noteworthy candidate. BODIPY, distinguished by its controllable molecular structure optical properties, outstanding fluorescence quantum yields, high molar absorption coefficients, remarkable chemical stability, undergone comprehensive investigation extensive exploration within realm of biological theranostics. This work aims to provide summary advancements development design strategies NIR‐II BODIPY fluorophores tailored advanced phototheranostics. Initially, elucidates several representative strategies, concluding electron‐programming strategy, extension conjugated backbone, J‐aggregation strategic establishment activatable fluorophores, which enhance skeletons. Subsequently, developments fluorescent BODIPY‐based nanoplatforms applications are intricately elaborated. In conclusion, this outlines future efforts directions refining meet evolving clinical demands. Acute kidney injuries (AKI) have serious short-term or long-term complications with high morbidity and mortality rate, thus posing great health threats. Developing high-performance NIR-II probes for noninvasive in situ detection of AKI via fluorescent optoacoustic dual-mode imaging is significance. Yet chromophores often feature long conjugation hydrophobicity, which prevent them from being renal clearable, limiting their applications the diseases. To fully exploit advantageous features heptamethine cyanine dye, while overcoming its relatively poor photostability, to strive design a probe imaging, herein, we developed PEG3-HC-PB, water soluble, biomarker activatable has good photostability. As probe, fluorescence (900-1200 nm) quenched due existence electron-pulling phenylboronic group (responsive element), it exhibits weak absorption peak at 830 nm. Meanwhile, presence overexpressed H2O2 region case AKI, converted phenylhydroxy group, enhances emission (600-900 eventually produces conspicuous signals imaging. This enables contrast-agent-induced ischemia/reperfusion-induced mice using real-time 3D-MSOT response H2O2. Hence, this can be used as practicable tool detecting AKI; additionally, strategy could provide insight into other large-conjugation multifarious biological applications.

Abstract Sudden hemorrhage stemming from internal organ wounds poses a grave and potentially fatal risk if left untreated. Injectable‐hydrogel‐based tissue sealants featuring multiple actions, including fit‐to‐shape in situ gelation, rapid hemostasis, pro‐angiogenic, anti‐bacterial outcome tracking, are ideal for the management of trauma wounds. Herein, an injectable‐hydrogel sealant AN@CD‐PEG&TQ which consists four‐arm 4‐arm poly(ethylene glycol) (PEG‐SC) succinimidyl carbonate), AN@CD nanoprobe, two bioactive peptides (anti‐microbial peptide Tet213 pro‐angiogenic QK) is developed. Among them, nanoparticles form through host/guest complexation amino‐group‐containing β‐cyclodextrin adamantyl group, enabling biomarker (NO)‐activatable optoacoustic/NIR‐II: Near‐infrared second biological window fluorescent imaging. The ample ─NH2 groups on surface readily engage cross‐linking with ester located at ends PEG‐SC. This expedites gelation process without necessitating additional initiators or agents; thus, significantly enhancing both hydrogel's application convenience biocompatibility. Bioactive (Tet213 safeguard against possible bacterial infections, facilitate angiogenesis, eventually, improve healing. hydrogel‐based demonstrates superior therapeutic bioimaging performance various mouse models liver hemorrhage, gastric perforation, bacterial‐infected skin wound models, highlighting its potential as high‐performance bleeding management.

Dyes with extended conjugate structures are the focus of extensive design and synthesis efforts, aiming to confer unique improved optical electronic properties. Such advancements render these dyes applicable across a wide spectrum uses, ranging from second-window near-infrared (NIR-II) bioimaging organic photovoltaics. Nevertheless, inherent benefits long conjugation often accompanied by persistent challenges like aggregation, fluorescence quenching, absorption blueshift, low stability poor water solubility. Herein, structural strategy termed "homo-dyad outer hydration layer" is introduced address problems, tailored for development imaging probes exhibiting absorption/emission wavelengths. This approach involves bringing two heptamethine cyanines together through flexible linker, forming homo-dyad structure, while strategically attaching four polyethylene glycol (PEG9) chains terminal heterocycles. imparts excellent solubility, biocompatibility, enhanced chemical, photo-, spectral dyes. Utilizing this strategy, biomarker-activatable probe (HD-FL-4PEG9-N) NIR-II fluorescent 3D multispectral optoacoustic tomography developed, its effectiveness in disease visualization.
