Abstract Reactive oxygen species (ROS)‐induced endoplasmic reticulum (ER) stress in sonodynamic therapy (SDT) can elicit immunogenic cell death (ICD)‐initiated antitumor immunity for augmented sono‐immunotherapy. However, unsatisfactory activity and mediocre ER induction ability of sonosensitizers essentially restrict SDT efficacy ICD stimulation. Herein, a versatile ER‐targeting Iridium(III) nanosonosensitizer is developed as superior inducer boosted tumor An ingenious cholic acid (CA)‐functionalized sonosensitizer Ir‐CA well‐designed skillfully crosslinked with human serum albumin (HSA) to form HSA@Ir‐CA. With high stability, favorable tumor‐targeting ability, reduction‐responsiveness, HSA@Ir‐CA preferentially accumulates sites enhanced cellular uptake, followed by rapid disassembly responding intracellular reductive environment. Abstract Reducing the scavenging capacity of reactive oxygen species (ROS) and elevating ROS production are two primary goals developing novel sonosensitizers for sonodynamic therapy (SDT). Hence, ultrathin 2D Bi 2 MoO 6 –poly(ethylene glycol) nanoribbons (BMO NRs) designed as piezoelectric glutathione (GSH)‐enhanced SDT. As a novel noninvasive therapeutic modality combining low-intensity ultrasound and sonosensitizers, sonodynamic therapy (SDT) is promising for clinical translation due to its high tissue-penetrating capability treat deeper lesions intractable by photodynamic (PDT), which suffers from the major limitation of low tissue penetration depth light. The effectiveness feasibility SDT are regarded rely on not only development stable flexible apparatus, but also screening sonosensitizers with good specificity safety. To give an outlook equipment, key technologies discussed according five aspects including ultrasonic dose settings, sonosensitizer screening, tumor positioning, temperature monitoring, reactive oxygen species (ROS) detection. In addition, some state-of-the-art multifunctional equipment integrating diagnosis treatment accurate introduced. Further, overview provided small molecular sensitizers nano/microenhanced sensitizers. Several types nanomaterial-augmented in discussion, porphyrin-based nanomaterials, porphyrin-like inorganic organic-inorganic hybrid nanomaterials different strategies improve efficacy. There no doubt that rapid will be promoted advanced smart nanomaterial-based sonosensitizer, multidisciplinary collaboration.

The development of efficient organic sonosensitizers is crucial for sonodynamic therapy (SDT) in the field cancer treatment. Herein, a new strategy based on triarylboron-doped acenethiophene scaffolds presented. attachment boron to linear acenethiophenes lowers lowest unoccupied molecular orbital (LUMO) energy, resulting redshifted absorptions and emissions. After encapsulation with amphiphilic polymer DSPE-mPEG2000 , it found that nanostructured BAnTh-NPs BTeTh-NPs (nanoparticles BAnTh BTeTh) shows hydroxyl radical (• OH) generation under ultrasound (US) irradiation aqueous solution almost no phototoxicity, which can overcome shortcomings O2 -dependent SDT avoid potential cutaneous phototoxicity issue. In vitro vivo therapeutic results validate boron-doped as enable high efficiency low good biocompatibility, indicating boron-functionalization acenes promising toward SDT.

Abstract Immunotherapy has made remarkable strides in cancer therapy over the past decade. However, such emerging still suffers from low response rates and immune‐related adverse events. Various strategies have been developed to overcome these serious challenges. Therein, sonodynamic (SDT), as a non‐invasive treatment, received ever‐increasing attention especially treatment of deep‐seated tumors. Significantly, SDT can effectively induce immunogenic cell death trigger systemic anti‐tumor immune response, termed immunotherapy. The rapid development nanotechnology revolutionized effects with robust induction. As result, more innovative nanosonosensitizers synergistic modalities are established superior efficacy safe profile. Despite the great promises of sonodynamic therapy (SDT) in combination cancer therapy, its clinical applications are hindered by "always-on" pharmacological activities therapeutic agents and lack efficient sonosensitizers. Herein, development semiconducting polymers as sonosensitizers further sono-immunotherapeutic nanobodies (SPNAb ) for activatable sono-immunotherapy reported. Conjugation anti-CTLA-4 antibodies onto polymer nanoparticles through a 1 O2 -cleavable linker affords SPNAb with relatively low CTLA-4 binding affinity. Upon sono-irradiation, generates not only to elicit effect induce immunogenic cell death, but also release trigger situ checkpoint blockade. Such synergistic action mediated modulates tumoricidal function T-cell immunity promoting proliferation cytotoxic T lymphocytes depleting immunosuppressive regulatory cells, resulting effective tumor regression, metastasis inhibition, durable immunological memory, prevention relapse. Therefore, this study represents proof-of-concept strategy using precise spatiotemporal control over immunotherapy.

Recent progress in cuproptosis sheds light on the development of treatment approaches for advancing sonodynamic therapy (SDT) due to its unique cell death mechanism. Herein, we elaborately developed an intelligent cell-derived nanorobot (SonoCu), composed macrophage-membrane-camouflaged nanocarrier encapsulating copper-doped zeolitic imidazolate framework-8 (ZIF-8), perfluorocarbon, and sonosensitizer Ce6, synergistically triggering cuproptosis-augmented SDT. SonoCu not only improved tumor accumulation cancer-cell uptake through cell-membrane camouflaging but responded ultrasound stimuli enhance intratumor blood flow oxygen supply, which consequently overcame barriers activated cuproptosis. Importantly, SDT effectiveness could be further amplified by multiple mechanisms, including reactive species accumulation, proteotoxic stress, metabolic regulation, sensitized cancer death. Particularly, exhibited ultrasound-responsive cytotoxicity against cells healthy cells, endowing it with good biosafety. Therefore, present first anticancer combination cuproptosis, may inspire studies pursuing a rational multimodal strategy.
