Abstract Abnormal activation of the intestinal mucosal immune system, resulting from damage to barrier and extensive invasion by pathogens, contributes pathogenesis inflammatory bowel disease (IBD). Current first‐line treatments for IBD have limited efficacy significant side effects. An innovative H 2 S‐releasing montmorillonite nanoformulation (DPs@MMT) capable remodeling homeostasis, repairing barrier, modulating gut microbiota is developed electrostatically adsorbing diallyl trisulfide‐loaded peptide dendrimer nanogels (DATS@PDNs, abbreviated as DPs) onto (MMT) surface. Abstract Hydrogen sulfide (H 2 S) plays crucial inflammatory modulating roles, representing a promising candidate for anti‐inflammatory therapies. However, current H S delivery approaches lack sufficient specificity against response. Herein, regarding the overexpressed aminopeptidase N (APN) at inflammation sites, an APN‐activated self‐immolative carbonyl (COS)/H donor ( AlaCOS ) was developed response‐specific delivery. The compound showed sustained generation upon APN activation in presence of carbonic anhydrase (CA), and responsiveness could be well regulated by amino acid sequence. Due to delivery, provided potent capability, which further validated RNA sequencing. In vivo experiments on full‐thickness cutaneous wound murine model also strong promoting effect healing, mainly due regulation response . By introducing caged coumarin fluorophore molecular architecture, self‐reporting fluorescence generated accompanied with APN‐mediated COS/H release, achieved visualization vitro vivo. Excessive accumulation of reactive oxygen and nitrogen species (RONS) dysbiosis intestinal microbiota are pivotal symptoms for inflammatory bowel disease (IBD) its associated complications, such as fibrosis. This research introduces a probiotic inulin hydrogel loaded with polypyrrole (PPy) nanozymes antifibrotic drug pirfenidone (PFD) (PPy/PFD@Inulin gel) designed the concurrent amelioration IBD fibrotic complication. Upon oral administration, gel matrix could extend gastrointestinal residence time PPy PFD, facilitating efficient reduction pro-inflammatory cytokine levels enhancement epithelial barrier repair well suppression fibrosis through sustained RONS scavenging, modulation gut attenuation TGF-β/Smad signaling pathway to inhibit fibroblast proliferation. Notably, PPy/PFD@Inulin demonstrated significant prophylactic therapeutic efficacy in acute chronic colitis induced by dextran sodium sulfate (DSS) mouse models. Rheumatoid arthritis (RA) represents an insidious autoimmune inflammatory disorder that severely lowers the life quality by progressively destructing joint functions and eventually causing permanent disability, posing a serious public health problem. Here, advanced theranostic probe is introduced integrates activatable second near-infrared (NIR-II) fluorescence imaging for precise RA diagnosis with multi-pronged treatments. A novel molecular comprising long-wavelength aggregation-induced emission unit manganese carbonyl cage motif synthesized, which enables NIR-II activation concurrently releasing therapeutic carbon monoxide (CO) gas in inflamed microenvironment. This self-assembles into biocompatible nanoprobe, subsequently conjugated anti-IL-6R antibody to afford active-targeting ability of RA. The nanoprobe exhibits significant turn-on signal at lesion, enabling highly sensitive real-time monitoring. combination ROS scavenging, on-demand CO release, IL-6 signaling blockade results potent effect synergistic immunomodulation impact, significantly alleviating symptoms preventing destruction. research introduces paradigm development high-performance, strategies facilitate detection enhanced treatment RA-related diseases.

Abstract Inflammatory bowel disease (IBD) encompasses a collection of chronic inflammatory conditions impacting the gastrointestinal tract, with discernible global rise in prevalence and severity. The overabundance reactive oxygen species (ROS) intestinal environment leads disruption local redox homeostasis, perturbation gut microbiota, exacerbating IBD. Consequently, mitigating excess ROS preserving microbiota homeostasis have emerged as effective approaches for IBD management. In this study, triangular DNA origami nanostructure loaded siRNA targeting tumor necrosis factor α ( siTNF‐α tDON) is introduced purpose. This demonstrates efficacy eliminating within cells closely associated IBD, diminishing inflammation, ameliorating disorders evidenced an mouse model. Furthermore, investigation effects tDON on related signaling pathways, including NF‐κB, MAPK/ERK Keap1/Nrf2.
