Abstract Background Multimorbidity (MM) is generally defined as the presence of 2 or more chronic diseases in same patient and seems to be frequently associated with frailty poor quality life. However, complex interplay between MM functional status hospitalized older patients has not been fully elucidated so far. Here, we implemented a 2-step approach, combining cluster analysis association rule mining explore how patterns disease associations change function disability. Methods this retrospective cohort study included 3,366 discharged from acute care units Ancona Cosenza sites Italian National Institute on Aging (INRCA-IRCCS) 2011 2017. While childhood adversity (CA) is known to be associated with multiple adverse outcomes, its link dementia an area limited exploration and inconsistent agreement. The study aimed examine the longitudinal associations of CA incident all-cause quantify potential mediating pathways. Data from UK Biobank. CA, encompassing neglect abuse, was evaluated retrospectively by online mental health questionnaire. Physical performance, psychological factors, lifestyles, biological indicators assessed at baseline were considered mediators. Incident defined International Classification Diseases, Tenth Revision codes obtained through self-reported medical conditions, primary care, hospital admission, death registrations. Cox proportional hazard models applied estimate associations. Mediation analyses conducted on mediators their contribution. This cohort comprised 150 152 nondemented individuals (mean [SD] age, 55.9 [7.7] years) (2006-2010). Compared who did not experience those exposed any exhibited a 30.0% higher risk (hazard ratio = 1.300, 95% confidence interval [CI]: 1.129-1.496). Each additional 15.5% (95% CI: 8.8%-22.5%, pfor trend < .001) increased risks. Depression, smoking, low grip strength explained 8.7%, 2.4%, 0.9% associations, respectively. Biomarkers involving inflammation, erythrocytes, liver, kidney function mediated 0.6%-1.4%. revealed detrimental effects identified some mediators, namely depression, strength, several targeted biomarkers. Abstract Dementia refers to an umbrella phenotype of many different underlying pathologies with Alzheimer’s disease (AD) being the most common type. Neuropathological examination remains gold standard for accurate AD diagnosis, however, that we know about genetics is based on Genome-Wide Association Studies (GWAS) clinically defined AD. Such studies have identified multiple susceptibility variants a significant portion heritability unexplained and highlighting phenotypic genetic heterogeneity entity. Background The atherosclerotic cardiovascular disease (ASCVD) is associated with dementia. However, the risk factors of dementia in patients ASCVD remain unclear, necessitating development accurate prediction models. Background/Objective: Although the context, personal conditions, and caregivers’ abilities influence nutrition of older people with dementia, adequate parameters are not always applied to identify these conditions. The aim this study was characterize nutritional care needs dementia their caregivers. Method: This descriptive exploratory conducted in Colombia. An intentional sample included 73 elderly individuals described participants’ characteristics GCPC-UN-D survey. Their conditions include medical history, objective tests, interviews. Abstract Background Complex diseases may share portions of their polygenic architectures which can be leveraged to identify drug targets with low off-target potential or repurposable candidates. However, the literature lacks methods make these inferences at scale using publicly available data. Methods We introduce a Bayesian model estimate structure trait only gene-based association test statistics from GWAS summary data and returns gene-level posterior risk probabilities (PRPs).
