Non-alcoholic fatty liver disease (NAFLD) is a global health burden closely linked to insulin resistance, obesity, and type 2 diabetes. The complex pathophysiology of NAFLD involves multiple cellular pathways molecular factors. Nuclear receptors (NRs) have emerged as crucial regulators lipid metabolism inflammation in NAFLD, offering potential therapeutic targets for NAFLD. Targeting PPARs FXRs has shown promise ameliorating symptoms halting progression. However, further investigation needed address side effects personalize therapy approaches. This review summarizes the current understanding involvement NRs pathogenesis explores their potential. We discuss role several modulating homeostasis liver, including peroxisome proliferator-activated (PPARs), X (LXRs), farnesoid (FXRs), REV-ERB, hepatocyte nuclear factor 4α (HNF4α), constitutive androstane receptor (CAR) pregnane (PXR).The expanding knowledge offers new avenues targeted therapies, necessitating exploration novel treatment strategies optimization existing approaches combat this increasingly prevalent disease.

Metabolic factors play a critical role in the development of digestive system cancers (DSCs), and East Asia has highest incidence malignant tumors system. We performed two-sample Mendelian randomization analysis to explore associations between 19 metabolism-related lifestyle clinical risk DSCs, including esophageal, gastric, colorectal, hepatocellular, biliary tract, pancreatic cancer. The causal association was explored for all combinations each factor DSC. gathered information on instrumental variables (IVs) from various sources retrieved outcome Biobank Japan (BBJ). data were studies east Asian populations. Finally, 17,572 DSCs cases 195,745 controls included. Our found that genetically predicted alcohol drinking strong indicator gastric cancer (odds ratio (OR) = 0.95; 95% confidence interval (CI): 0.93-0.98) hepatocellular carcinoma (OR 1.11; CI: 1.05-1.18), whereas coffee consumption had potential protective effect 0.69; 0.53-0.90). Thyroid hormones (THs) have been found that it is closely associated with the onset and progression of non-alcoholic fatty liver disease (NAFLD). However, current study could not verify intrinsic relationship between thyroid NAFLD, which requires further research. The searches studies reported both TH level in serum NAFLD were performed PubMed, Web Science, Cochrane Library, Embase databases. We combined an overall meta-analysis a dose-response to assess correlation function levels risk NAFLD. Overall, 10 included total 38,425 individuals. non-linear model showed for every 1 ng/dL increase FT4, was reduced by 10.56% (p=0.003). odds ratios (ORs) high free triiodothyronine (FT3) exposure compared those low FT3 1.580 (95% CI 1.370 1.830, I2 = 0.0%, p<0.001) meta-analysis. continuous variable indicated individuals TSH (SMD=1.32, 95% 0.660 1.970, had significantly higher fibrosis than levels. Our findings only validate there occurrence abnormal THs, expected more observational will still be conducted future demonstrate Registered number PROSPERO: CRD42023405052.
