Rhein (RH), a natural anthraquinone compound, is considered an effective treatment candidate for ulcerative colitis (UC), whose multiple biological activities contribute to UC, including anti-inflammation, antioxidation, intestinal barrier repair, and microflora regulation. However, the application of RH severely limited by its low water solubility, bioavailability, poor colonic targeting. Although some nanoparticles have been developed oral delivery RH, most them mainly highlighted only one effect drug strategies but above activities. Therefore, polysaccharide-based nanodelivery system, comprising chitosan (CS) fucoidan (FU), with pH/reactive oxygen species (ROS) sensitivity mucosal adhesion, was first used load as comprehensive UC. Briefly, RH-F/C-NPs were prepared using polyelectrolyte self-assembly method; average size 233.1 ± 5.7 nm, encapsulation rate 93.67 1.60%. And it could maintain gastric stability release in colon designed pH/ROS contributed structures. Cellular uptake experiments showed that both NCM 460 cells RAW 264.7 had good RH-F/C-NPs. Importantly, effects vivo experiments, results which significantly reduce DSS-induced inflammation inhibiting TLR4/NF-κB-mediated anti-inflammatory pathway, Nrf2/HO-1-mediated antioxidant In addition, pharmacokinetic studies shown F/C-NPs increase plasma concentration accumulation extent. short, this study develop nanosystem dual "one stone four birds" therapeutic on

Background: An animal model of DSS-induced UC has been widely used in basic research, and the dysbiosis gut microbiome is one important pathogenetic mechanisms UC, but its dynamic changes correlation with inflammatory factors are not clear yet. Methods: Clinical signs tissue damage degree C57BL/6 ulcerative colitis mice induced by different concentrations DSS were compared that normal mice, finally optimal concentration was determined. Then we analyzed sequencing results to determine patterns their factors. Results: at 2.5% 3.0% could cause intestinal injury induce colitis. However, resulted higher mortality. In addition, there model: relative abundance flora increased first then decreased Bacteroides , Parabacteroides Romboutsia Clostridium_sensu_stricto_1, Lachnospiraceae_NK4A136_group, norank_f_norank_o_Clostridia_UCG-014, Parasutterella Lactobacillus Muribaculum norank_f_Muribaculaceae, Bifidobacterium Coriobacteriaceae_UCG-002 Enterorhabdus did change 14 days significantly on day 21. Moreover, cytokines closely associated imbalance microbiota UC: most pathogenic bacteria tract positively correlated pro-inflammatory negatively anti-inflammatory factors, while beneficial opposite. Conclusion: Intestinal microecology plays an role model, dynamically occurrence development Keywords: colitis, DSS, microbiome, changes, 16S rDNA

Increasing evidence suggests that the gut microbiota may represent potential strategies for Parkinson’s disease (PD) treatment. Our previous research revealed a decreased abundance of Akkermansia muciniphila (Akk) in PD mice; however, whether Akk is beneficial to unknown. To answer this question, mice received MPTP intraperitoneally construct subacute model and were then supplemented with orally 21 consecutive days. Motor function, dopaminergic neurons, neuroinflammation, neurogenesis examined. In addition, intestinal inflammation, serum fecal short-chain fatty acids (SCFAs) analyses, assessed. We found treatment effectively inhibited reduction neurons substantia nigra pars compacta (SNpc) partially improved motor function mice. Additionally, markedly alleviated neuroinflammation striatum hippocampus promoted hippocampal neurogenesis. It also level colon inflammation. Furthermore, these aforementioned changes are mainly accompanied by alterations isovaleric acid levels, lower permeability. strongly neuroprotective agent therapy.

Ulcerative colitis (UC) is a chronic inflammatory disease with uncontrolled inflammation and demage to the intestinal barrier. Rhein, bioactive compound in traditional Chinese medicine, has anti-inflammatory repair effect. However, their clinical application limited by hydrophobicity poor bioavailability. L-arginine, as complement NO, synergistic attenuating effects. In this paper, red/NIR-I fluorescent carbon dots based on rhein doped L-arginine (RA-CDs), which are synthesized hydrothermal process without any organic solvents, reported. RA-CDs preserve portion of functional group active precursor, increase solubility, emit light for biological imaging. vitro experiments show that scavenge excessive reactive oxygen species (ROS), protect cells from oxidative stress, enable fluorescence imaging inflamed colons. DSS-induced UC mouse model, both delayed prophylactic treatment via intraperitoneal tail vein injections alleviate severity reducing restoring This study highlights novel strategy treating poorly soluble small-molecule drugs.
