Background: Alzheimer’s disease (AD) is a neurodegenerative characterized by beta-amyloid (Aβ) deposition and increased acetylcholinesterase (AchE) enzyme activities. Indole 3 carbinol (I3C) diindolylmethane (DIM) are reported to have neuroprotective activities against various neurological diseases, including ischemic stroke, Parkinson’s disease, neonatal asphyxia, depression, stress, neuroinflammation, excitotoxicity, except for AD. In the present study, we investigated anti-AD effects of I3C DIM. Methods: Docking molecular dynamic studies AchE network pharmacological were conducted DIM’s self AchE-induced Aβ aggregation investigated. The DIM Aβ-induced neurotoxicity assessed in SH-S5Y5 cells observing cell viability ROS. Results: with show binding efficiency −7.0 −10.3, respectively, dynamics revealed better interaction stability between AchE. Network indicated that interacted several proteins involved pathophysiology Further, significantly inhibited (IC50: (18.98 µM) (11.84 µM)) self-induced aggregation. Both compounds enhanced exposed reduced equipotential neuroprotection when compared donepezil. Conclusions: Our findings indicate both inhibiting induced

Brain-derived neurotrophic factor (BDNF) and its downstream tropomyosin receptor kinase B (TrkB) signaling pathway play pivotal roles in the resilience action of antidepressant drugs, making them prominent targets psychiatric research. Oxidative stress (OS) contributes to various neurological disorders, including neurodegenerative diseases, stroke, mental illnesses, exacerbates aging process. The nuclear erythroid 2-related 2 (Nrf2)-antioxidant responsive element (ARE) serves as primary cellular defense mechanism against OS-induced brain damage. Thus, Nrf2 activation may confer endogenous neuroprotection OS-related damage; notably, TrkB/phosphoinositide 3-kinase (PI3K)/protein (Akt) pathway, stimulated by BDNF-dependent TrkB signaling, activates promotes translocation. However, insufficient neurotrophin support often leads downregulation diseases. targeting Nrf2-ARE system is a promising therapeutic strategy for treating Phytochemicals, indole-3-carbinol (I3C) metabolite, diindolylmethane (DIM), exhibit neuroprotective effects through BDNF’s mimetic activity; Akt phosphorylation induced, antioxidant activated blocking Nrf2-kelch-like ECH-associated protein 1 (Keap1) complex. This review emphasizes potential I3C derivatives concurrently activating neuronal mechanisms treatment

Neuroinflammation in response to environmental stressors is an important common pathway a number of neurological and psychiatric disorders. Responses immune-mediated stress can lead epigenetic changes the development neuropsychiatric Isothiocyanates (ITC) have shown promise combating oxidative inflammation nervous system as well organ systems. While sulforaphane from broccoli most widely studied ITC for biomedical applications, their precursor glucosinolates are found many species cruciferous other vegetables including moringa. In this review, we examine both clinical pre-clinical studies on amelioration disorders (neurodevelopmental, neurodegenerative, other) 2018 present, documentation protocols several ongoing studies. During time, there been 16 (9 randomized controlled trials), which reported effect autism spectrum disorder schizophrenia. We also review over 80 preclinical examining treatment brain-related dysfunctions The evidence date reveals great potential treating these conditions with minimal toxicity. authors call well-designed trials further translation potent phytochemicals into therapeutic practice.

3,3'-Diindolylmethane (DIM), a naturally occurring bis-indole found in cruciferous vegetables and produced small amounts the normal flora of human gut, has demonstrated neuroprotective benefits models CNS hypoxia stroke. In CNS, DIM modulates activation aryl hydrocarbon receptor (AhR) inhibits its pro-inflammatory effects. Although capable crossing blood brain barrier, DIM's bioavailability is limited by low solubility. Dispersed BR4044 provides nanoscale high-solubility suspension with potential for treating traumatic injury (TBI). The present study aimed to determine whether treatment could reduce pathology improve behavioral recovery following moderate TBI. Male Sprague Dawley rats received fluid percussion or sham surgery followed vehicle acute period. TBI animals showed significantly reduced cortical hippocampal edema lower levels serum-derived extracellular vesicles compared Vehicle animals. preserved sensorimotor function associative fear memory. Cortical contusion size neuronal loss parietal cortex CA3 region hippocampus were also treatment. decreased microbleeding nuclear AhR at site. This translational demonstrates that ameliorates improves neurological outcomes reducing edema, lowering vesicle release, modulating AhR, preserving neurons, red cell (RBC) extravasation into injured brain, promoting recovery.
