Wound healing is a complex, variable, and time-dynamic repair process. Wounds can be classified as acute wounds or chronic wounds, effective wound management still major challenge in clinical nursing settings. The microenvironment collectively regulated by internal biomolecules, external drugs, sanitation. Traditional dressings (powders, bandages, sponges, etc.) often have poor therapeutic effects during processes because they cannot respond to the dynamic changes over long-term. Stimulus-responsive biomaterials, which are activated various factors intrinsic influences, hold great promise for precise drug delivery controlled release. Various stimulus-responsive hydrogels been developed recent years, exhibiting range of “smart” properties, such interacting with wound, sensing conditions environmental changes, responding accordingly, thereby effectively promoting healing. Several natural Chinese herbal medicines have demonstrated considerable potential in facilitating wound healing, while the primary concern remains centered around optimizing formulation and structure to maximize their efficacy. To address this, a microneedles drug delivery system is proposed that harnesses gelatinized starch key ingredients-aloe vera berberine. After aged well-designed mold, starch-based are fabricated with suitable mechanical strength load components. The resulting hydrogel microneedles, enriched integrated berberine aloe, exhibit antibacterial, anti-inflammatory, fibroblast growth-promoting properties, thereby healing whole process. The conventional oral administration of pioglitazone for Type II diabetes management is frequently compromised by hepatic first-pass metabolism and associated systemic adverse effects, necessitating the development enhanced transdermal delivery approaches. This study developed a drug system combining conductive hydrogel microneedles iontophoresis to improve hydrochloride (PIO) its therapeutic efficacy in treatment type diabetes. microneedles, fabricated using poly(methyl vinyl ether-alt-maleic anhydride) as main matrix material, exhibited excellent conductivity, mechanical strength, high loading capacity. In vitro permeation experiments demonstrated that, when combined with at current intensity 0.5 mA, cumulative PIO reached 238.1 ± 27.14 μg/cm2 within 48 h, significantly higher than that microneedle group alone. diabetic rat model, microneedle-iontophoresis displayed better hypoglycemic effect group, blood glucose reduction 6.3 mmol/L on day 8, 5.1 positive control group. Pharmacokinetic analysis indicated Tmax, T1/2, mean residence time were longer those administration, indicating sustained-release characteristics. By leveraging swelling hydrogels, nanomaterial integration, stimuli-responsive properties, HMNs provide precision medicine capabilities across diverse therapeutic diagnostic applications. However, challenges remain in mechanical stability, hydrogel-based MNs must balance flexibility with sufficient strength skin penetration.
