Perfluorooctane sulfonate (PFOS) has been reported to induce hepatotoxicity in wildlife and humans. Novel PFOS alternatives have widely used following restrictions on PFOS, but little is known about their potential toxicity. Here, the first comprehensive investigation chronic underlying molecular mechanisms of 6:2Cl-PFESA (F-53B), sodium p-perfluorous nonenoxybenzene (OBS) was carried out adult zebrafish through a histopathological examination, biochemical measurement, multi-omics analysis. its caused changes liver histopathology function indices order F-53B > OBS, which consistent with concentration liver. In silico modeling transcriptional profiles suggested that aberrant hepatic lipid metabolism induced by initiated action peroxisome proliferator-activated receptor γ (PPARγ), triggered downstream genes transcription led an imbalance between synthesis expenditure. Gut microbiome analysis provided another novel mechanistic perspective abundance Legionella, Ralstonia, Brevundimonas, Alphaproteobacteria, Plesiomonas, Hyphomicrobium might link alterations PPAR pathway based significant correlation. This study provides insight into highlights need for concern environmental exposure risks.

Due to its special flavour and cheapness, starch is a source of nutrition for humans most animals, some whom even prefer consume large amounts starchy foods. However, the use by carnivorous fish limited excessive intake can lead liver damage, but mechanism damage not clear. Therefore, in this study, two isonitrogenous isolipid semi-pure diets, Z diet (0% starch) G (22% starch), were formulated, respectively. The largemouth bass ( M . salmoides ) cultured fiberglass tanks randomly divided into groups fed diets 45 days. Blood collected on day 30 enzymology, histopathology, ultramicropathology, flow cytometry, transcriptomics investigate high mechanism. results showed that affect growth performance bass. caused whitening an increase hepatopancreas index (HSI), aspartate aminotransferase (AST), alanine (ALT) serum. Histopathological observations led severe vacuolisation, congestion, moderate necrotizing hepatitis liver. significant postprandial blood glucose insulin serum bass, promoting synthesis accumulation hepatic glycogen liver, leading loss hepatocyte organelles inducing fibrosis. Meanwhile, induced production oxidative stress promoted apoptosis necrosis hepatocytes. Transcriptome analysis revealed there 10,927 2,656 unique genes groups, KEGG enrichment 19 pathways significantly enriched, including those related metabolism cell survival. Network mapping based differential expressing emergence regulatory network dominated PI3K/Akt signaling pathway.
