Physical activity has a positive effect on human health and emotional well-being. However, in both amateur professional athletes, training poses risk of acute or chronic injury through repetitive overloading bones, joints, muscles. Inflammation can be an adverse intense exercise caused by several factors including oxidative stress. The present narrative review summarizes current knowledge inflammatory markers induced physical exercise. Post-exercise recovery may reduce responses is key to effective adaptation muscle tissues sustained exertion.

Lysosomes are organelles involved in cell metabolism, waste degradation, and cellular material circulation. They play a key role the maintenance of physiological homeostasis. Compared with lysosomal content other organs, that kidney is abundant, abnormalities associated occurrence development certain renal diseases. Lysosomal structure function intrinsic cells impaired diabetic disease (DKD). Promoting biosynthesis and/or restoring can repair damaged podocytes proximal tubular epithelial cells, delay progression DKD. homeostasis may be advantageous alleviating Here, we systematically reviewed latest advances relationship between dyshomeostasis DKD based on recent literature to further elucidate mechanism injury diabetes mellitus highlight application potential as new prevention treatment strategy for However, research screening effective interventions still its infancy, thus should focus future studies. The out cell-specific regulation targets according different stages DKD, so realize controllable targeted during successful clinical this therapeutic strategy.

Diabetic kidney disease (DKD) is a major microvascular complication of diabetes and leading cause end-stage renal worldwide. Autophagy plays an important role in maintaining cellular homeostasis physiology. In DKD, the accumulation advanced glycation end products induces decreased autophagy-related protein expression transcription factor EB (TFEB) nuclear transfer, to impaired autophagy lysosomal function blockage autophagic flux. This accelerates resident cell injury apoptosis, mediates macrophage infiltration phenotypic changes, ultimately aggravated proteinuria fibrosis DKD. Natural polyphenols show promise treating DKD by regulating promoting transfer TFEB repair. review summarizes characteristics potential application mechanisms some known natural as regulators with goal contributing deeper understanding polyphenol treatment development their applications. Finally, we point out limitations current research provide outlook for future research.

Abstract Kidney filtration is ensured by the interaction of podocytes, endothelial and mesangial cells. Immunoglobulin accumulation at barrier pathognomonic for glomerular injury. The mechanisms that regulate filter permeability are unknown. Here, we identify a pivotal role proteasome in specific cell type. Combining genetic inhibitor-based human, pig, mouse, Drosophila models demonstrate maintains integrity, with podocytes requiring constitutive cells immunoproteasomal activity. Endothelial immunoproteasome deficiency as well inhibition disrupt mice, resulting pathologic immunoglobulin deposition. Mechanistically, observe reduced endocytic activity, which leads to altered membrane recycling receptor turnover. This work expands concept (immuno)proteasome control protease orchestrating protein degradation antigen presentation endocytosis, providing new therapeutic targets treat disease-associated accumulations.

Protein homeostasis strongly depends on the targeted and selective removal of unneeded or flawed proteins, protein aggregates, damaged excess organelles by two main intracellular degradative systems, namely ubiquitin proteasomal system (UPS) autophagosomal lysosomal system. Despite representing completely distinct mechanisms degradation, which underlie differing regulatory mechanisms, growing evidence suggests that UPS autophagy interact especially in situations overwhelming impairment, both are involved podocyte proteostasis pathogenesis injury. The differential impact biology disease development progression is not understood. Recent advances understanding role reviewed here.

Fabry disease (FD) is an X-linked lysosomal caused by variants of the GLA gene; formation defective alpha-galactosidase A contributes to accumulation substrates in several organs. Chronic inflammation thought contribute organ damage FD patients. In total, 36 classic patients (15 men/21 women) and 25 healthy controls (20 men/8 were assessed. The Mainz Severity Score Index (MSSI) was established after conducting interviews with chart review. Serum IL-6, IL-1β, TNF-α levels evaluated both groups. The mean age (years) for 43.1±15.4 that 47.4±12.2 (p>0.05). Twenty-two (59.5%) treated enzyme replacement therapy (ERT). IL-6 significantly higher than controls. Patients ERT had serum those not ERT. There no difference IL-1β between who not. MSSI scores correlated (r=0.60, p<0.001) (r=0.45, p<0.001).FD associated elevated this cohort. ERT, particularly, women, exhibited ERT; reflecting greater burden.
