Abstract Colorectal cancer (CRC) is a malignant tumor originating from epithelial cells of the colon or rectum, and its invasion metastasis could be regulated by anoikis. However, key genes pathways regulating anoikis in CRC are still unclear require further research. The single cell transcriptome dataset GSE221575 GEO database was downloaded applied to subpopulation type identification, intercellular communication, pseudo time trajectory analysis, receptor ligand expression analysis CRC. Meanwhile, RNA TCGA, GSE39582, GSE17536, GSE17537 datasets were merged into one bulk dataset. differentially expressed (DEGs) related extracted these data sets, marker obtained after feature selection. Abstract Peritoneal metastasis is a critical step in the progression of gastric cancer (GC), yet its underlying mechanisms remain poorly understood. Here, we identify FERMT2, member Kindlin protein family, as key regulator anoikis resistance (AR) and peritoneal GC. FERMT2 expression increases suspension-time-dependent manner associated with higher pathological grade, advanced clinical stage, poorer prognosis. Functional studies vitro vivo demonstrate that promotes AR facilitates metastasis. Mechanistically, suppresses ubiquitination SOX2, thereby enhancing stability up-regulating FN1 transcription. Furthermore, report TGFβ-RI also manner, forming positive feedback loop via TGFβ-1/TGFβ-RI signaling. This drives extracellular fibronectin matrix deposition, strengthens cell–matrix interactions, supports AR.
