Circulating tumor cells (CTCs) are pivotal in cancer metastasis and serve as valuable biomarkers for diagnosis, prognosis, treatment monitoring. Traditional CTC capture methods predominantly utilize the epithelial cell adhesion molecule (EpCAM) a marker isolation. However, heterogeneity of these circulating epithelial-to-mesenchymal transition process (wherein acquire mesenchymal characteristics) limit efficacy EpCAM-based techniques. In this paper, we critically review role EpCAM capture, explore impact on expression, discuss alternative strategies to enhance By evaluating limitations EpCAM-mediated challenges posed by transition, aim provide insights into development more comprehensive liquid biopsy approaches management.

Abstract Cancer treatment faces many hurdles and resistance is one among them. Anti-cancer strategies are evolving due to innate acquired capacity, governed by genetic, epigenetic, proteomic, metabolic, or microenvironmental cues that ultimately enable selected cancer cells survive progress under unfavorable conditions. Although the mechanism of drug being widely studied generate new target-based drugs with better potency than existing ones. However, broader flexibility in resistance, advanced therapeutic options efficacy need be explored. Combination therapy an alternative a success rate though risk amplified side effects commonplace. Cancer stem cells (CSCs) have emerged as key contributors to tumor initiation, growth, and metastasis. In addition, CSCs play a significant role in inducing immune evasion, thereby compromising the effectiveness of cancer treatments. The reciprocal communication between microenvironment (TME) is observed, with TME providing supportive niche for CSC survival self-renewal, while CSCs, turn, influence polarization persistence TME, promoting an immunosuppressive state. Consequently, these interactions hinder efficacy current therapies, necessitating exploration novel therapeutic approaches modulate target CSCs. this review, we highlight intricate strategies employed by evade surveillance develop resistance therapies. Furthermore, examine dynamic interplay shedding light on how interaction impacts progression. Moreover, provide overview advanced that specifically which hold promise future clinical translational studies treatment.

The extracellular matrix (ECM) governs a wide spectrum of cellular fate processes, with particular emphasis on anoikis, an integrin-dependent form cell death. Currently, anoikis is defined as intrinsic apoptosis. In contrast to traditional apoptosis and necroptosis, integrin correlates ECM signaling intracellular cascades, describing the full process anoikis. However, frequently overlooked in physiological pathological processes well vitro research models. this review, we summarized role spanning embryonic development, organ tissue repair, inflammatory responses, cardiovascular diseases, tumor metastasis, so on. Published: March 1, 2025

Abstract Peritoneal metastasis is a critical step in the progression of gastric cancer (GC), yet its underlying mechanisms remain poorly understood. Here, we identify FERMT2, member Kindlin protein family, as key regulator anoikis resistance (AR) and peritoneal GC. FERMT2 expression increases suspension-time-dependent manner associated with higher pathological grade, advanced clinical stage, poorer prognosis. Functional studies vitro vivo demonstrate that promotes AR facilitates metastasis. Mechanistically, suppresses ubiquitination SOX2, thereby enhancing stability up-regulating FN1 transcription. Furthermore, report TGFβ-RI also manner, forming positive feedback loop via TGFβ-1/TGFβ-RI signaling. This drives extracellular fibronectin matrix deposition, strengthens cell–matrix interactions, supports AR. These findings establish pivotal mediator GC, offering insights into potential therapeutic target.
