Background Neoadjuvant systemic therapy has been shown to benefit patients with solid tumors such as breast cancer and colorectal cancer, but its application in hepatocellular carcinoma (HCC) is still the exploratory stage, no established effective regimen. This systematic review meta-analysis aims investigate efficacy safety of neoadjuvant resectable HCC. Methods The clinical trials HCC PubMed, Embase Cochrane Library were systematically searched. A was performed using STATA/MP18.0 software, effect size calculated either a fixed effects model or random model, 95% confidence intervals (CIs) calculated. Subgroup analysis according Results included 328 from 15 studies. In HCC, pooled pathologic complete response (pCR) rate 15% (95%CI: 10%–21%), major (MPR) 28% 21%–35%), incidence grade 3–4 treatment-related adverse events (TRAEs) 11% (95% CI: 4%–20%), objective (ORR) 27% 20%–35%), surgical resection 84% 75%–92%), delay 0.00% 0%–4%). results subgroup showed that targeted combined immunotherapy superior dual ICI (immune checkpoint inhibitor) combination monotherapy, while monotherapy highest, ICIs immunotherapy. Conclusion shows preliminarily beneficial outcomes treatment. However, future large-scale multicenter randomized controlled are needed confirm this conclusion. Cancer poses intricate challenges to treatment due its complexity and diversity. Ferroptosis circular RNAs (circRNAs) are emerging as innovative therapeutic avenues amid the evolving landscape of cancer therapy. Extensive investigations into circRNAs reveal their diverse roles, ranging from molecular regulators pivotal influencers ferroptosis in cell lines. The results underscore significance modulating pathways that impact crucial aspects development, including survival, proliferation, metastasis. A detailed analysis delineates these pathways, shedding light on mechanisms through which influence ferroptosis. Building upon recent experimental findings, study evaluates potential targeting induce By identifying specific associated with etiology cancer, this paves way for development targeted therapeutics exploit vulnerabilities cells. This review consolidates existing understanding circRNAs, emphasizing role therapy providing impetus ongoing research dynamic field. See also graphical abstract(Fig. 1).

With the increasing impact of hepatocellular carcinoma (HCC) on society, there is an urgent need to propose new HCC diagnostic biomarkers and identification models. Histone lysine lactylation (Kla) affects prognosis cancer patients emerging target in treatment. However, potential Kla-related genes poorly understood. A variety machine learning methods were used construct validate a model differentially expressed Kla with comprehensive evaluations included ROC, Kaplan‒Meier curve, Cox regression, decision curve. Immune infiltration gathered spatial transcriptome was performed using integrated data from multiple databases. Furthermore, single-cell analysis discover cell–cell communication Mendelian randomization study causal relationships between immune cell HCC. Lastly, qRT-PCR verify expression genes. We established consisting 12 that had well prognostic performance identified as independent factors. Single-cell showed CD8 T+ cells conventional dendritic enriched patients. Spatial transcriptomics indicated influenced characteristics results TBNK monocytes main risk validation Huh7 significantly higher than LO2 cells. Overall, established, which may provide strategies insights for treatment diagnosis

Epidermal growth factor receptor substrate 15 (EPS15) is part of the EGFR pathway and has been implicated in various tumorigenesis.Increasing evidence suggests that long noncoding RNA (lncRNA) plays an essential role liver hepatocellular carcinoma (LIHC) by regulating expression proteins genes.Through analysis cancer genome atlas (TCGA) database, we found EPS15 highly expressed LIHC tissue, lncRNA EPS15-antisense1 (EPS15-AS1) decreased cell lines. However, function EPS15-AS1 still unknown. When was overexpressed HepG2 lines, reduced activity invasiveness were inhibited. In addition, observed increase Fe 2+ ion lipid peroxidation after overexpression EPS15-AS1, further showed susceptibility to ferroptosis increased.Aldo-keto reductase family 1 member B (AKR1B1) belongs aldo/keto superfamily involved maintaining cellular redox balance.Survival revealed patients with a higher level AKR1B1 have lower survival rate TCGA database. We also enhanced LIHC, had ability promote invasiveness. Moreover, alleviated promoting effect on ferroptosis. Therefore, can induce cells inhibiting disrupting balance.EPS15 may serve as biomarkers for diagnosis potential drug LIHC.

Tumor-associated macrophages (TAMs) are pivotal in the tumor microenvironment (TME) of hepatocellular carcinoma (HCC), influencing various stages from initiation to metastasis. Understanding role TAMs HCC is crucial for developing novel therapeutic strategies. Macrophages exhibit plasticity, resulting M1 and M2 phenotypes, with displaying antitumor properties promoting progression. Targeting alter their polarization could offer new avenues treatment. β,β-dimethylacrylalkannin (DMAKN), a natural naphthoquinone, has gained attention its properties. However, impact on modulation remains unclear. This study investigates DMAKN's anti-HCC activity. Using an vitro model THP-1 cells, we induced LPS/IFN-γ IL-4/IL-13, confirming specific markers. Co-culturing these cells showed that inhibited growth, while promoted it. Screening non-toxic DMAKN concentrations revealed ability induce enhance LPS/IFN-γ-induced macrophages, both showing effects. Conversely, suppressed IL-4/IL-13-induced polarization, inhibiting macrophages' promotion cell viability. In summary, induces enhances thereby growth. These findings suggest potential regulate HCC, offering promise future development.
