The synovial tissues under rheumatoid arthritis conditions are usually infiltrated by inflammatory cells, particularly M1 macrophages with aberrant redox homeostasis, which causes rapid deterioration of articular structure and function. Herein, we created an ROS-responsive micelle (HA@RH-CeOX) through the in situ host–guest complexation between ceria oxide nanozymes hyaluronic acid biopolymers, precisely delivered nanozyme clinically approved drug Rhein (RH) to proinflammatory macrophage populations inflamed tissues. abundant cellular ROS could cleave thioketal linker trigger release RH Ce. Specifically, Ce3+/Ce4+ pair present SOD-like enzymatic activity rapidly decompose alleviate oxidative stress macrophages, while inhibit TLR4 signaling both act a concerted manner induce their repolarization into anti-inflammatory M2 phenotype ameliorate local inflammation promote cartilage repair. Notably, rats bearing showed drastic increase M1-to-M2 ratio from 1:0.48 1:1.91 tissue significantly reduced cytokine levels including TNF-α IL-6 following intra-articular injection HA@RH-CeOX, accompanied efficient regeneration restored Overall, this study revealed approach modulate homeostasis reprogram polarization states micelle-complexed biomimetic enzymes, offers alternative opportunities for treatment arthritis.

Abstract Osteoarthritis (OA), a chronic degenerative joint disease, is highly prevalent among the aging population, and often leads to pain, disability, diminished quality of life. Although considerable research has been conducted, precise molecular mechanisms propelling OA pathogenesis continue be elusive, thereby impeding development effective therapeutics. Notably, recent studies have revealed subchondral bone lesions precede cartilage degeneration in early stage OA. This marked by escalated osteoclast‐mediated resorption, subsequent imbalances metabolism, accelerated turnover, decrease volume, contributing significantly pathological changes. While role hallmarks extensively elucidated from perspective chondrocytes, their connection with osteoclasts not yet fully understood. There compelling evidence suggest that age‐related abnormalities such as epigenetic alterations, proteostasis network disruption, cellular senescence, mitochondrial dysfunction, can stimulate osteoclast activity. review intends systematically discuss how contribute pathogenesis, placing particular emphasis on age‐induced shifts It also aims future probing into therapeutic approaches targeting during aging.

Objectives The association between the neutrophil percentage to albumin ratio (NPAR) and risk of osteoarthritis (OA) rheumatoid arthritis (RA) remains unclear. This study aims investigate NPAR OA RA. Methods cross-sectional analyzed data from 92,062 American adults in NHANES database 1999 2016. Various statistical analyses were conducted associations risks RA, including multivariable logistic regression, subgroup analysis, smooth curve fitting, threshold effect analysis. Results After screening, final population included 36,147 participants, with 3,881 individuals diagnosed 2,178 adjusting for confounding factors, higher levels associated an increased RA (OR=1.05; 95% CI: 1.03-1.07; P <0.0001), but not (OR=1.01; 0.99-1.02; =0.755). was remarkably consistent across subgroups by age, sex, body mass index (BMI), alcohol consumption, hypertension, diabetes, smoking status. Further using fitting models revealed a nonlinear inflection point identified at 15.56. Conclusion High is positively prevalence provides us new insights management treatment patients.

Osteoarthritis (OA) is a degenerative disease closely related to aging. Nevertheless, the role and mechanisms of aging in osteoarthritis remain unclear. This study aims identify potential aging-related biomarkers OA explore genes immune microenvironment synovial tissue.Normal gene expression profile microarrays were obtained from Gene Expression Omnibus (GEO) database (ARGs) Human Aging Genomic Resources (HAGR). Ontology (GO), Kyoto Encyclopedia Genes Genomes (KEGG), Disease (DO), set variation analysis (GSVA) enrichment used uncover underlying mechanisms. To Hub ARDEGs with highly correlated features (Hub OA-ARDEGs), Weighted Co-expression Network Analysis (WGCNA) machine learning methods used. Furthermore, we created diagnostic nomograms receiver operating characteristic curves (ROC) assess OA-ARDEGs' ability diagnose predict which miRNAs TFs they might act on. The Single sample (ssGSEA) algorithm was applied look at infiltration characteristics their relationship OA-ARDEGs.We discovered 87 normal synovium samples. According functional enrichment, are primarily associated inflammatory regulation, cellular stress response, cell cycle transcriptional regulation. OA-ARDEGs excellent identified as MCL1, SIK1, JUND, NFKBIA, JUN. Wilcox test showed that all significantly downregulated validated validation by qRT-PCR. Using ssGSEA algorithm, 15 types six activation increased samples well OA-ARDEGs.Synovial may promote progression inducing inflammation. JUN can be novel biomolecular markers therapeutic targets for OA.

Osteoarthritis (OA) is one of the most prevalent musculoskeletal disorders globally, and treating OA remains a significant challenge. Currently, pharmacological treatments primarily aim to alleviate symptoms associated with inflammation pain, no disease-modifying therapies are available delay development progression. Reactive oxygen species (ROS) play an essential role in progression, which promising target for curing OA. In this study, it was found that photothermal properties near-infrared (NIR) irradiation enhanced ROS scavenging activity molybdenum-based polyoxometalate (POM) nanoclusters. Because scavenging, NIR-responsive POM nanoclusters were developed as novel excellent nano-antioxidants protection. The results demonstrated exhibited outstanding antioxidant superexcellent anti-inflammatory effects, could effectively clinical mice, diminish inflammatory cytokines, reduce catabolic proteases, mitigate progression Meanwhile, local treatment had side effects on normal tissues. Thus, study pioneered application alleviating expected safety efficiency.

Abstract Background Chronic inflammation may contribute to increased mortality risk in individuals with osteoarthritis (OA), but research on the prognostic value of inflammatory biomarkers is limited. We aimed evaluate associations systemic immune–inflammation index (SII) and response (SIRI) all-cause cardiovascular among US adults OA. Methods This cohort study included 3545 OA aged ≥ 20 years from National Health Nutrition Examination Survey 1999–2020. The SII SIRI were calculated using complete blood cell count data. Participants categorized as having a higher or lower cutoff points derived by maximally selected rank statistics method. Cox proportional hazards models, Fine–Gray competing regression models time-dependent receiver operating characteristic (ROC) analysis used between SII/SIRI patients. Results Over median follow-up 5.08 (3.42–9.92) years, 636 (17.94%) deaths occurred, including 149 (4.20%) deaths. According multivariable-adjusted involving demographic, socioeconomic, health factors, patients had twofold greater than (HR 2.01; 95% CI: 1.50–2.68). Similarly, was associated an 86% relative 1.86; 1.46–2.38). Similar trend found mortality, elevated 88% 67% respectively, compared 1.88; 1.16–3.03) 1.67; 1.14–2.44).
