Cancer management faces multiple obstacles, including resistance to current therapeutic approaches. In the face of challenging microenvironments, cancer cells adapt metabolically maintain their supply energy and precursor molecules for biosynthesis thus sustain rapid proliferation tumor growth. Among various metabolic adaptations observed in cells, altered glucose metabolism is most widely studied. The aberrant glycolytic modification has been associated with cell division, growth, progression, drug resistance. higher rates glycolysis as a hallmark modulated by transcription factor hypoxia inducible 1 alpha (HIF-1α), downstream target PI3K/Akt signaling, deregulated pathway cancer. We provide detailed overview current, primarily experimental, evidence on potential effectiveness flavonoids combat glycolysis-induced conventional targeted therapies. manuscript focuses reducing via affecting PI3K/Akt, HIF-1α (as critical that regulated pathway), key mediators PI3K/Akt/HIF-1α signaling (glucose transporters enzymes). working hypothesis proposes – an attractive application mitigate Phytochemicals represent source promising substances applicable primary, secondary, tertiary care. Reprogramming of cancer metabolism has become increasingly concerned over the last decade, particularly reprogramming glucose metabolism, also known as "Warburg effect". The is considered a novel hallmark human cancers. A growing number studies have shown that can regulate many biological processes cancers, including carcinogenesis, progression, metastasis, and drug resistance. In this review, we summarize major functions, clinical significance, potential targets signaling pathways metabolic in Moreover, applications natural products small molecule inhibitors targeting are analyzed, some agents trial statuses summarized, well pros cons for therapy analyzed. Overall, plays an important role prediction, prevention, diagnosis treatment Glucose reprogramming-related great to serve biomarkers improving individual outcomes prognosis patients. innovations related will be hotspot research future. We suggest more high-quality trials with abundant formulations toxicology experiments would beneficial development application drugs metabolism. This review provide researchers broader perspective comprehensive understanding about significance

Prostate cancer (PCa) remains one of the clinically relevant pathologies that needs pragmatic and effective treatment approaches. The current study aimed to evaluate anticancer potential epigallocatechin gallate (EGCG) EGCG-loaded nanoparticles (EGCG NP) for prostate in in-vitro 3D spheroid model. EGCG NPs was synthesized by using polymeric method as reported our previous study. A assay conducted human specific cell lines (PC3 22Rv1) cultured on poly-HEMA-covered plates at different time points. Once formed, spheroids were treated with either alone or continuously 6 days. Simultaneously, controls also taken comparison purpose. CellROX dye used quantitate formation reactive oxygen species (ROS) response 22Rv1 PC3, spheroids. evaluated measure modulation mitochondrial membrane quantification apoptotic live cells a flow cytometer. sizes both studied found be significantly (p < 0.05) reduced after its nanoformulation. significant increase ROS observed PC3 EGCG-NPs treatment. However, no change cells. Similarly, compounds NPs) did not show any changes Interestingly, showed between but nanoformulation didn't number Our doses highlighting possible advantage model specifically further preclinical vivo studies are recommended suitable decipher vitro data exploit against cancer. results indicate efficacy PCa lines.

Cancer causes many deaths worldwide each year, especially due to tumor heterogeneity leading disease progression and treatment failure. Targeted of heterogeneous population cells - cancer stem is still an issue in protecting affected individuals against associated multidrug resistance progression. Nanotherapeutic agents have the potential go beyond state-of-the-art approaches overall management. Specially assembled nanoparticles act as carriers for targeted drug delivery. Several nanodrugs already been approved by US Food Drug Administration (FDA) treating different types. Phytochemicals isolated from plants demonstrate considerable nanomedical applications oncology thanks their antioxidant, anti-inflammatory, anti-proliferative, other health benefits. Phytochemical-based NPs can enhance anticancer therapeutic effects, improve cellular uptake agents, mitigate side effects toxic treatments. Per evidence, phytochemical-based specifically target CSCs decreasing risks relapse metastatic manifestation. Therefore, this review focuses on current outlook targeting research studies consideration framework predictive, preventive, personalized medicine (3PM).

Apocynin (Apo), an NADPH oxidase (NOX) inhibitor, has been widely used to treat various inflammatory diseases. However, the therapeutic effects of Apo on benign prostatic hyperplasia (BPH), a multifactorial disease associated with chronic inflammation and hormone imbalance, remain unknown. The link between androgen signaling, reactive oxygen species (ROS), prostate cell proliferation may contribute pathogenesis BPH; therefore, aim this study was identify specific signaling pathway involved demonstrate whether anti-oxidant plays role in prevention treatment BPH. Ingenuity analysis si-RNA transfection were conducted receptor (AR) NOX4 linkage Pathological markers BPH measured by H&E staining, immunoblotting, ELISA, qRT-PCR, immunofluorescence examine effect Apo. Abstract With the increase of aging population and prevalence obesity, incidence cardiovascular disease (CVD) cancer has also presented an increasing tendency. These two different diseases, which share some common risk factors. Relevant studies in field reversing Cardio-Oncology have shown that phenotype CVD a significant adverse effect on tumor prognosis, is mainly manifested by positive correlation between malignant progression concomitant tumors. This distal crosstalk link diseases makes us aware importance diagnosis, prediction, management personalized treatment systemic diseases. The circulatory system bridges interaction cancer, suggests we need to fully consider holistic characteristics these process clinical treatment. circulating exosome-miRNAs been intrinsically associated with -related regulation, become one focuses basic research (as biomarker). changes expression profiles disease-associated miRNAs (Cardio-miRNAs) may adversely affect In this article, sorted screened tumor-related miRNA data based literature, then summarized their commonalities (several important pathways), further discussed conclusions related experimental studies. We take approach considering as factor for malignancy, provides in-depth analysis various regulatory mechanisms or pathways involved dual attribute (Cardio-/Onco-miRNAs). will be key revealing effects tumors highlight nature Therefore, Cardio-miRNAs should given great attention from researchers tumors, might new targets Meanwhile, principles precision medicine (such predictive preventive medicine, 3PM) reverse Cardio-oncology better improve individual outcomes, developing therapy perspective protecting function.

Micro ribonucleic acids (microRNAs or miRNAs) form a distinct subtype of non-coding RNA and are widely recognized as one the most significant gene expression regulators in mammalian cells. Mechanistically, regulation occurs through microRNA binding with its response elements 3’-untranslated region target messenger RNAs (mRNAs), resulting post-transcriptional silencing genes, expressing mRNAs. Compared to small interfering RNAs, microRNAs have more complex regulatory patterns, making them suitable for fine-tuning expressions different tissues. Dysregulation is well known causative factors malignant cell growth. Today, there numerous data points regarding cancer transcriptomes, specificity changes various tissues, predictive value specific biomarkers.
