Abstract Advances in DNA metabarcoding have greatly expanded our knowledge of microbial communities recent years. Pipelines and parameters been tested extensively for bacterial using the 16S rRNA gene best practices are largely established. For fungal ITS gene, however, only a few studies considered how such pipelines can affect community prediction. Here we report novel bias uncovered during ITS2 sequencing Trichoderma -infected ant fungus gardens confirmed mock communities. Abnormally low forward read quality caused reads to be computationally filtered before pair merging, thus almost entirely eliminating ASVs from resulting profiles. Saprobic fungi are known for their critical role in decomposition and nutrient cycling. The study of saprobic is equally important, as it helps understanding ecological roles identifying hidden diversity. This focused on Aquilaria , which poorly studied compared to economically important hosts like coffee, tea, rubber. Our rigorous process led the collection two new terrestrial from Guangdong Yunnan provinces China. Fungal infections are a significant global health challenge, causing approximately 3.8 million deaths annually, with immunocompromised populations particularly at risk. Traditional antifungal therapies, including azoles, echinocandins, and polyenes, face limitations due to rising resistance, toxicity, inadequate treatment options. This review explores innovative strategies for preventing managing fungal infections, such as vaccines, peptides, nanotechnology, probiotics, immunotherapy. Vaccines offer promising avenues long-term protection, despite difficulties in their development complexity immune evasion mechanisms. Antifungal peptides provide novel class of agents broad-spectrum activity reduced resistance risk, whilst nanotechnology enables targeted, effective drug delivery systems. Probiotics show potential vulvovaginal candidiasis, by maintaining microbial balance. Immunotherapy leverages system modulation enhance defenses, omics technologies deliver comprehensive insights into biology, paving the way therapeutic vaccine targets. While these approaches hold immense promise, challenges cost, accessibility, translational barriers remain. A coordinated effort among researchers, clinicians, policymakers is critical advancing addressing burden effectively.

Four new fungi, viz. Clavulinopsiswumengshanensis (Clavariaceae, Agaricales), Henningsomycesbambusae (Porotheleaceae, Xenasmabisterigmatae, and X.guttulata (Xenasmataceae, Russulales), from Yunnan Province in China, are proposed, based on a combination of morphological features molecular evidence. Phylogenetic analyses were conducted using combined dataset internal transcribed space nuclear ribosomal RNA large subunit sequences. In the present study, five new wood-inhabiting fungal species, Conferticiumalbocremeum (Stereaceae, Russulales), Dendrocorticiopsisluteoalba (Punctulariaceae, Corticiales), Eichleriellabiluoxueshanensis (Auriculariaceae, Auriculariales), Gloeohypochniciumyunnanense (Incertae sedis, and Punctularianigrodontea collected from southern China, are proposed based on a combination of morphological features phylogenetic evidence. is characterized by membranaceous smooth basidiomata with white to cream surface ellipsoid verrucose basidiospores (9-11 × 5-7 µm); buff, ellipsoid, thin-walled (7-8 4.5-5.5 coriaceous grandinioid buff cinnamon-buff hymenophore allantoid, (13.5-17.5 7-9 cream, globose, thick-walled warted (10-12 10-11.5 µm), resupinate effused-reflexed (8.5-10 5-6 µm). Sequences internal transcribed spacers (ITS) large subunit (nLSU) nuclear ribosomal DNA (rDNA) markers studied samples were generated. Wood-inhabiting fungi play an integral role in wood degradation and the cycle of matter ecological system. They are considered a ʻkey playerʼ decomposition because their ability to produce all kinds enzymes that break down lignin, cellulose hemicellulose. In this study, some wood-inhabiting mushrooms were collected from Yunnan-Guizhou Plateau, Asia, based on morphological molecular evidence, they introduced as Punctochaete murina J.H.Dong & C.L.Zhao gen. et sp. nov. is characterized by resupinate basidiomata with punctate hymenial surface, monomitic hyphal system generative hyphae bearing clamp connections, cylindrical allantoid, thin-walled, smooth, slightly curved basidiospores. Sequences internal transcribed spacer (ITS) large subunit (nLSU) markers studied samples generated, phylogenetic analyses performed maximum likelihood, parsimony Bayesian inference methods. Phylogenetic ITS+nLSU nuclear RNA gene regions showed forms monophyletic lineage within order Auriculariales which it sister clade comprising two closely related genera: Exidiopsis Tremellochaete.

Russulales comprises a highly diverse group of species with respect to basidiomata morphology and hymenophore configuration, in which this order is heterogeneous, that can be classified as resupinate, effused-reflexed, discoid, clavarioid, pileate, or stipitate, varied hymenophores such smooth, hydnoid, poroid, lamellate different russuloid species. Species have been considered significant economic value. Six new wood-inhabiting fungi belonging the genera Conferticium , Gloeocystidiellum Peniophora two families, Peniophoraceae Stereaceae (Russulales), were found southwest China. Sequences ITS+nLSU loci six taxa generated, phylogenetic analyses performed maximum likelihood, parsimony, Bayesian inference methods an emphasis on phylogeny smooth order. The combined analysis showed grouped within Russulales, tuberculatum cremeum G. fissuratum into family Stereaceae, albohymenia P. hengduanensis punctata Peniophoraceae. multi-gene confirmed novelty placement taxa.
