Mental disorders and neurological diseases have brought rapidly increasing medical burdens. Although extensive researches been conducted hugely, effective therapies still progressing slowly. Current dilemma reminds us that the human is a superorganism. Only if we take self its partner microbiota into consideration at same time can better understand these diseases. Over last few centuries, has experienced tremendous changes much more than own genes because of modern transformations in diet, lifestyle, care, so on, paralleling epidemiological transition. Existing research indicates gut plays an important role this According to gut–brain psychology, crucial part gut–brain, it communicates with brain via microbiota–gut–brain axis. The almost develops synchronously brain, mind. Gut influences various normal mental processes phenomena they are involved pathophysiology numerous Targeting therapy for promising approach supported by three theories: hypothesis, "old friend" leaky theory. effects behavior fulfilled through microbiota-gut-brain axis which mainly composed nervous pathway, endocrine immune pathway. Undoubtedly, psychology will bring great enhancement neuroscience, psychiatry. Various improving methods including FMT, probiotics, prebiotics, healthy lifestyle shown capability promote function brain. Harnessing improve health, prevent related be true future.

Diet its bioactive components have recognized among modifiable risk factors, possibly influencing their pathogenesis. This review aimed to summarize molecular mechanisms underlying putative beneficial effects toward brain different dietary such as micro- macronutrient intake habits, feeding time circadian rhythm. The role hormonal homeostasis context glucose metabolism adiponectin regulation impact on systemic neuro-inflammation also considered deepened. In addition, effect individual molecules exerting antioxidant activities acting anti-inflammatory agents, omega-3 fatty acids polyphenols, for central nervous system via modulation adult neurogenesis, synaptic neuronal plasticity, microglia activation summarized. Current medication for anxiety disorders is suboptimal in terms of efficiency and tolerability, highlighting the need improved drug treatments. In this review an overview drugs being studied different phases clinical trials their potential treatment fear-, anxiety- trauma-related presented. One strategy followed development refining improving compounds interacting with existing anxiolytic targets, such as serotonergic prototypical GABAergic benzodiazepines. A more innovative approach involves search novel mechanisms action using growing knowledge base concerning relevant neurocircuitries neurobiological underlying pathological fear anxiety. The target systems evaluated include glutamate, endocannabinoid neuropeptide systems, well ion channels targets derived from phytochemicals. Examples promising candidates currently generalised disorder, social panic obsessive compulsive disorder or post-traumatic stress ketamine, riluzole, xenon one common pharmacological modulation glutamatergic neurotransmission, neurosteroid aloradine. Finally, D-cycloserine, MDMA, L-DOPA cannabinoids have shown efficacy enhancing fear-extinction learning humans. They are thus investigated augmentative speeding up long-term effectiveness exposure-based psychotherapy, which could render chronic dispensable many patients. These efforts indicative a rekindled interest renewed optimism discovery field, after decades relative stagnation.

Aging is characterized by the functional decline of tissues and organs increased risk aging-associated disorders, which pose major societal challenges are a public health priority. Despite extensive human genetics studies, limited progress has been made linking with aging. There growing realization that altered assembly, structure dynamics gut microbiota actively participate in aging process. Age-related microbial dysbiosis involved reshaping immune responses during aging, manifest as immunosenescence (insufficiency) inflammaging (over-reaction) accompany many age-associated enteric extraenteric diseases. The can be regulated, suggesting potential target for interventions. This review summarizes recent findings on physiological succession across life-cycle, roles mechanisms healthy alterations diseases, microbiota-targeted anti-aging strategies.

Abstract Fibromyalgia (FM) is a prevalent syndrome, characterised by chronic widespread pain, fatigue, and impaired sleep, that challenging to diagnose difficult treat. The microbiomes of 77 women with FM 79 control participants were compared using 16S rRNA gene amplification whole-genome sequencing. When comparing patients unrelated controls differential abundance analysis, significant differences revealed in several bacterial taxa. Variance the composition was explained FM-related variables more than any other innate or environmental variable correlated clinical indices FM. In line observed alteration butyrate-metabolising species, targeted serum metabolite analysis verified levels butyrate propionate patients. Using machine-learning algorithms, microbiome alone allowed for classification (receiver operating characteristic area under curve 87.8%). To best our knowledge, this first demonstration gut nonvisceral pain. This observation paves way further studies, elucidating pathophysiology FM, developing diagnostic aids possibly allowing new treatment modalities be explored.
