To overcome negative adverse effects and improve therapeutic index of dexamethasone (Dex) in rheumatoid arthritis (RA), we developed a novel sustained release formulation-intra-articular injectable dexamethasone-loaded thermosensitive hydrogel (DLTH) with chitosan-glycerin-borax as carrier for the remission inflammation pain. The focus this article is to explore both anti-inflammatory pain-relieving DLTH joint injection bovine type-II collagen-induced (CIA) rats.Wistar rats were randomized into three groups, including normal group (n=6), model (n=6) (n=10). Joint (1mg/kg Dex per rat) was injected on day 12 twice week weeks. Clinical signs body weight, paw swelling scores, histologic analysis, hind mechanical withdrawal threshold (MWT), plantar pressure pain (PPT) taken consideration. Serum contents IL-17A, prostaglandin E2 (PGE2), prostacyclin 2 (PGI2) D2 (PGD2), real-time polymerase chain reaction (PCR) analysis inflammatory factors pain-related mediators synovium dorsal root ganglia (DRG), Western blotting NF-κB all evaluated.Paw swelling, scores destruction attenuated DLTH-treated group. Results showed that not only down-regulated serum but also mRNA levels NGF, key proteins pathway synovium. Increases MWT PPT elucidated pain-reducing DLTH. Elevated PGD2 declines PGE2 PGI2, genes DRGs recorded.These data impeded synovial processes through down-regulating transcription activity pathway, intra-articular may aid regulation RA regulating conduction process.

The circadian rhythm (CR) is a fundamental biological process regulated by the Earth’s rotation and solar cycles. It plays critical role in various bodily functions, its dysregulation can have systemic effects. These effects impact metabolism, redox homeostasis, cell cycle regulation, gut microbiota, cognition, immune response. Immune mediators, proteins, hormones exhibit oscillations, supporting optimal function defence against pathogens. Sleep deprivation disruptions challenge regulatory mechanisms, making responses vulnerable. Altered CR pathways been implicated diseases such as diabetes, neurological conditions, autoimmune (SADs). SADs involve abnormal to self-antigens, with genetic environmental factors disrupting self-tolerance contributing conditions like Systemic Lupus Erythematosus, Rheumatoid Arthritis, Inflammatory Myositis.
