Abstract Bovine male fertility in animals has a direct impact on the productivity of dairy herds. The epididymal sperm maturations involve extensive surface modifications to gain fertilizing ability, especially by absorptions plethora biomolecules, including glycoprotein beta-defensins (BDs), enzymes, organic ions, protein, and phospholipids. Defensins are broad-range nonspecific antimicrobial peptides that exhibit strong relations with innate adaptive immunity, but their roles relatively recently identified. In course evolution, BD genes give rise different clusters specific functions, reproductive undergoing duplications nonsynonymous mutations. polymorphisms have been reported milk compositions, disease resistance, activities. However, recent decades, link emerged as an appealing improvement performance such motility, membrane integrity, cervical mucus penetration, evading uterus immunosurveillance, oviduct cell attachment, egg recognition. reproductive-specific glycosylated class-A BDs (CA-BDs) shown age- sex-specific expressions organs, signifying physiological pleiotropism, maturation transport female tract. By considering adult organ-specific expressions, importance functionalities, bioinformatic analysis, we selected two bovine BBD126 BBD129 novel potential biomarkers fertility. Despite BDs, however, genomic characterization most across livestock nonmodel organisms remains predictive/incomplete. current review discusses our understanding pleiotropic polymorphism, structural attributes concerning fertilizability gamete animals.

Abstract This single cell study aimed to clarify whether an elevated incidence of sperm with a retained cytoplasmic droplet (CD) can be compensated by higher number in boar semen doses maintain fertility. Cluster analysis motile spermatozoa (ten boars) revealed that CD are underrepresented the fast, linearly moving cohort compared morphologically normal sperm. Nonetheless, response motility stimulator procaine was barely affected distal (Cramer’s V = 0.14), but moderately proximal (V 0.28). Viability lower ( p < 0.05) not during 168 h storage extended samples (n 11) and subsequent thermic stress. Morphologically from normospermic 10) or high (≥ 15%) 9) had similar patterns responses procaine. The origin no effect on viability > 0.05; n 26). In conclusion, enhanced prevalence seems compensable increase numbers doses.

African swine fever (ASF) has evolved from an exotic animal disease to a threat global pig production. An important avenue for the wide-spread transmission of diseases is their dissemination through boar semen used artificial insemination. In this context, we investigated role male reproductive organs in ASF. Mature domestic boars and adolescent wild boars, inoculated with different ASF virus strains, were by means virological pathological methods. Additionally, electron microscopy was employed investigate vitro sperm. However, some other cell types, including Leydig, endothelial stromal cells, also positive. Using RNAScope, p72 mRNA scattered halo epididymal duct epithelium, which point disruption barrier. No direct infection spermatozoa observed immunohistochemistry, or microscopy. Taken together, our results strengthen assumption that ASFV can transmitted via semen. Future studies are needed explore excretion dynamics efficiency.

The main cation/calcium channel of spermatozoa (CatSper), first identified in 2001, has been thoroughly studied to elucidate its composition and function, while distribution among species sperm sources is yet incomplete. CatSper composed several subunits that build a pore-forming calcium channel, mainly activated vivo ejaculated cells by intracellular alkalinization progesterone, as suggested the vitro examinations. relevance dual: maintain homeostasis (alongside plethora membrane channels present) well being involved pre-fertilization events, such capacitation, hyperactivation motility acrosome reaction, with remarkable differences. Abstract Background The endoplasmic reticulum (ER) is the central hub for protein quality control, where disulfide isomerases (PDIs), encoded by at least 21 genes, play a pivotal role. These multifunctional proteins contribute to bond formation, proper folding, and modifications, may act as hormone‐binding (e.g., steroids), influencing hormone biology. interplay between ER proteostasis, PDIs, epididymis—a crucial site sperm maturation—remains largely understudied. Objectives This study characterizes transcriptional signatures of Pdi genes in epididymis. Material methods Transcriptional profiles selected were assessed adult Wistar rat tissues, epididymis under different experimental conditions (developmental stages, surgical castration, efferent ductules ligation [EDL]). In silico bioinformatic analyses identified expression trends this gene family human epididymal segments.
