The blood-brain barrier (BBB) plays a vital role in protecting the central nervous system (CNS) by preventing entry of harmful pathogens from bloodstream. However, this also presents significant obstacle when it comes to delivering drugs for treatment neurodegenerative diseases and brain cancer. Recent breakthroughs nanotechnology have paved way creation wide range nanoparticles (NPs) that can serve as carriers diagnosis therapy. Regarding their promising properties, organic NPs potential be used effective drug delivery across BBB based on recent advancements. These remarkable ability penetrate using various mechanisms. This review offers comprehensive examination intricate structure distinct properties BBB, emphasizing its crucial function preserving balance regulating transport ions molecules. disruption conditions such stroke, Alzheimer's disease, Parkinson's disease highlights importance developing creative approaches drugs. Through encapsulation therapeutic molecules precise targeting processes vasculature, NP formulations present hopeful strategy improve BBB. We explore changes pathological investigate factors affect successful into brain. In addition, we most systems associated with shown positive results treating ischemic disorders. opens up new possibilities nanotechnology-based therapies cerebral diseases.

The blood-brain barrier (BBB) is a crucial structure that maintains brain homeostasis by regulating the entry of molecules and cells from bloodstream into central nervous system (CNS). Neurodegenerative diseases such as Alzheimer's Parkinson's disease, well ischemic stroke, compromise integrity BBB. This leads to increased permeability infiltration harmful substances, thereby accelerating neurodegeneration. In this review, we explore mechanisms underlying BBB disruption, including oxidative stress, neuroinflammation, vascular dysfunction, loss tight junction integrity, in patients with neurodegenerative diseases. We discuss how breakdown contributes neurotoxicity, abnormal accumulation pathological proteins, all which exacerbate neuronal damage facilitate disease progression. Furthermore, potential therapeutic strategies aimed at preserving or restoring function, anti-inflammatory treatments, antioxidant therapies, approaches enhance integrity. Given role neurodegeneration, maintaining its represents promising approach slow prevent progression

The aim of this study was to investigate the antioxidant capacity fruit wines and their protective effects against hydrogen peroxide-induced oxidative stress in rat synaptosomes vitro. were produced from strawberries drupe fruits (i.e., plum, sweet cherry, peach, apricot) through microvinification with a pure S. cerevisiae yeast culture. Fruit without added sugar before start fermentation, whereas subvariants pits only applied wines. First, treated wines, while induced H2O2. Subsequently, activities enzymes (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx)) content malondialdehyde (MDA), an indicator membrane injury, determined. In addition, Briggs–Rauscher reaction (BR) used evaluate inhibition free radicals. All investigated increased activity studied decreased MDA compared corresponding controls (synaptosomes H2O2). After synaptosomal treatment plum wine, highest observed for SOD (5.57 U/mg protein) GPx (0.015 protein). Strawberry wine CAT (0.047 showed best ability reduce lipid peroxidation, yielding lowest level (2.68 nmol/mg). Strawberry, cherry identified as samples higher both principal component analysis (PCA) hierarchical cluster (HCA). Finally, exhibited inhibitory BR (397 s). results suggest that could be considered potential functional food due stress.

Oxidative stress (OS) is an established hallmark of cancer and neurodegenerative disorders (NDDs), which contributes to genomic instability neuronal loss. This review explores the contrasting role OS in stem cells (CSCs) NDDs. Elevated levels reactive oxygen species (ROS) contribute promote tumor initiation progression CSCs, while NDDs such as Alzheimer’s Parkinson’s disease, accelerates death impairs cellular repair mechanisms. Both scenarios involve disruption delicate balance between pro-oxidant antioxidant systems, leads chronic oxidative stress. Notably, CSCs neurons display alterations redox-sensitive signaling pathways, including Nrf2 NF-κB, influence cell survival, proliferation, differentiation. Mitochondrial dynamics further illustrate these differences: enhanced function supports adaptability whereas impairments heighten vulnerability. Understanding common mechanisms OS-induced redox imbalance may provide insights for developing interventions, addressing aging hallmarks, potentially mitigating or preventing both

Diet is an important nonpharmacological risk-modifying factor for Alzheimer's disease (AD). The approaches used here to assess diet's role in the risk of AD include multi-country ecological studies, prospective and cross-sectional observational laboratory studies. Ecological studies have identified fat, meat, obesity from high-energy diets as factors reported that rates peak about 15-20 years after national dietary changes. Observational compared Western pattern with those Dietary Approaches Stop Hypertension (DASH), Mediterranean (MedDi), Mediterranean-DASH Intervention Neurodegenerative Delay (MIND) diets. Those including higher consumption saturated total fats, ultraprocessed foods a lower fruits, legumes, nuts, omega-3 fatty acids, vegetables, whole grains. Diet-induced associated significant inflammation, insulin resistance, oxidative stress, elevated homocysteine, advanced glycation end products, trimethylamine N-oxide. molecular mechanisms by which bioactive components specific affect are discussed. Given most countries' entrenched food supply systems, upward trends would be hard reverse. However, people willing able, low-animal product diet plenty anti-inflammatory, low-glycemic load may helpful.
