DNA serves as the foundation for molecular biology, leading to development of numerous techniques. Often, these techniques necessitate separation and visualization specific regions. Electrophoresis provides a solution this requirement. However, purification from agarose gels presents significant challenge, both in terms complexity cost. Therefore, here we propose two main protocols that are cost-effective efficient based on silica columns or freezing follow by alcohol precipitation. In case column extraction, gel was partially completely dissolved, yielding most situations. extraction precipitation with ethanol, obtained only out three treatments. A successful bacterial transformation PCR were achieved confirmed suitability recovered further applications.

Multiplex Polymerase Chain Reaction (PCR) has significantly impacted the field of infectious disease diagnostics, offering rapid and precise identification bacterial fungal pathogens. Unlike traditional culture methods, which may take days to yield results, multiplex PCR provides diagnostic insights within hours, enabling faster, targeted antimicrobial therapy reducing delay in treating critical infections like sepsis. The technique’s high sensitivity broad pathogen coverage make it ideal for both single polymicrobial infections, improving outcomes across respiratory, bloodstream, bacterial/fungal infections. However, is not without challenges; initial costs need specialized training can limit its adoption, especially low-resource settings. This review discusses clinical advantages limitations PCR, highlighting influence on accuracy, stewardship, global fight against resistance (AMR). Furthermore, recent innovations such as digital portable devices, are explored potential tools expanding access diagnostics worldwide.

This method offers streamlined time-efficient procedure contrast to the widely used next-generation sequencing Studies have reported encouraging results dPCR detecting fetal autosomal aneuploidies. Consequently, this systematic review aimed evaluate effectiveness of screening trisomy 21, 18, 13. Methods A search was conducted PubMed, Web Sciences, Embase relevant articles published up December 30, 2023. The Quality Assessment Diagnostic Accuracy Studies-2 (QUADAS-2) utilized quality assessment included articles. Furthermore, bivariate random-effect regression model conduct meta-analysis on utility 21 screening. Results total 9 were review, with all them assessing screening, 2 1 studies conducting additional analysis abilities 18 13, respectively. random-effects calculated pooled sensitivity specificity 95% confidence interval (CI). Meta-analysis 6 comparing trisomy-21 karyotyping demonstrated dPCR's 98% [95% CI: 94 -100] 99% 99 -100]. While 13 proved impractical, values favorable. Conclusions These findings suggest that holds promise as an effective tool testing, presenting less time-consuming intricate alternative sequencing.
