Metabolic syndrome is a cluster of conditions associated with the risk diabetes mellitus type 2 and cardiovascular diseases (CVDs). closely related to obesity. Increased adiposity promotes inflammation oxidative stress, which are precursors various complications involving metabolic components, namely insulin resistance, hypertension, hyperlipidemia. An increasing number studies confirm importance stress chronic in etiology syndrome. However, few have reviewed mechanisms underlying role contributing In this review, we highlight by reactive oxygen species (ROS) increase mitochondrial dysfunction, protein damage, lipid peroxidation, impair antioxidant function Biomarkers can be used disease diagnosis evaluation severity.

Insulin resistance (IR) plays a crucial role in the development and progression of metabolism-related diseases such as diabetes, hypertension, tumors, nonalcoholic fatty liver disease, provides basis for common understanding these chronic diseases. In this study, we provide systematic review causes, mechanisms, treatments IR. The pathogenesis IR depends on genetics, obesity, age, drug effects. Mechanistically, any factor leading to abnormalities insulin signaling pathway leads host, including receptor abnormalities, disturbances internal environment (regarding inflammation, hypoxia, lipotoxicity, immunity), metabolic function organelles, other abnormalities. available therapeutic strategies are mainly exercise dietary habit improvement, chemotherapy based biguanides glucagon-like peptide-1, traditional Chinese medicine (e.g., herbs acupuncture) can also be helpful. Based current there still some vacancies follow up consider, is need define more precise biomarkers different lifestyle interventions, explore natural or synthetic drugs targeting treatment. This could enable treatment patients with multiple combined diseases, aim treating disease holistically reduce healthcare expenditures improve quality life extent.

The occurrence of obesity has increased across the whole world. Many epidemiological studies have indicated that strongly contributes to development cancer, cardiovascular diseases, type 2 diabetes, liver diseases and other disorders, accounting for a heavy burden on public health-care systems every year. Excess energy uptake induces adipocyte hypertrophy, hyperplasia formation visceral fat in non-adipose tissues evoke disease, diseases. Adipose tissue can also secrete adipokines inflammatory cytokines affect local microenvironment, induce insulin resistance, hyperglycemia, activate associated signaling pathways. This further exacerbates progression obesity-associated Although some progress treatment been achieved preclinical clinical studies, pathogenesis obesity-induced are complex unclear. We still need understand their links better guide In this review, we review between with view improve future management its co-morbidities.

Nowadays, diabetes mellitus has emerged as a significant global public health concern with remarkable increase in its prevalence. This review article focuses on the definition of and classification into different types, including type 1 (idiopathic fulminant), 2 diabetes, gestational hybrid forms, slowly evolving immune-mediated ketosis-prone other special types. Diagnostic criteria for are also discussed. The role inflammation both is explored, along mediators potential anti-inflammatory treatments. Furthermore, involvement various organs highlighted, such adipose tissue obesity, gut microbiota, pancreatic β-cells. manifestation Langerhans β-cell islet inflammation, oxidative stress, impaired insulin production secretion addressed. Additionally, impact liver cirrhosis, acute kidney injury, immune system complications, diabetic complications like retinopathy neuropathy examined. Abstract Although aging has traditionally been viewed as the most important risk factor for osteoarthritis (OA), an increasing amount of epidemiological evidence highlighted association between metabolic abnormalities and OA, particularly in younger individuals. Metabolic abnormalities, such obesity type II diabetes, are strongly linked to they affect both weight-bearing non-weight-bearing joints, thus suggesting that pathogenesis OA is more complicated than mechanical stress induced by overweight. This review aims explore recent advances research on relationship risk, including impact abnormal glucose lipid metabolism, potential targeted therapeutic strategies.

Abstract Sarcopenia is a multifactorial disease characterized by reduced muscle mass and function, leading to disability, death, other diseases. Recently, the prevalence of sarcopenia increased considerably, posing serious threat health worldwide. However, no clear international consensus has been reached regarding etiology sarcopenia. Several studies have shown that insulin resistance may be an important mechanism in pathogenesis induced attenuation that, conversely, can lead resistance. causal relationship between two not clear. Abstract Obesity, which is the accumulation of fat in adipose tissue, has adverse impacts on human health. Obesity-related metabolic dysregulation similarities to alterations observed aging. It been shown that adipocytes obese individuals undergo cellular aging, known as senescence. Senescence can be transmitted other normal cells through a series chemical factors referred senescence-associated secretory phenotype (SASP). Most these are pro-inflammatory compounds. The immune system removes senescent T-cells, but immunosenescence, senescence cells, disrupts clearance T-cells. Immunosenescence occurs result aging or indirectly transmission from tissues. significant occurrence obesity expected cause immunosenescence and impairs response resolve inflammation. sustained chronic inflammation insulin's actions Therefore, this review focuses role adipocyte obesity-associated subsequent dysregulation. Moreover, article suggests novel therapeutic approaches improve syndrome by targeting T-cells using senotherapeutics. Graphical

Abstract Background Obesity has increased in recent years with consequences on diabetes and other comorbidities. Thus, 1 out of 3 diabetic patients suffers cardiovascular disease (CVD). The network among glucose, immune system, endothelium epicardial fat an important role pro-inflammatory thrombotic mechanisms atherogenesis. Since semaglutide, long-acting glucagon like peptide 1- receptor agonist (GLP-1-RA), a glucose-lowering drug, reduces body weight, we aimed to study its effects human (EAT), aortic endothelial cells neutrophils as atherogenesis involved-cardiovascular cells. Methods EAT subcutaneous (SAT) were collected from undergoing cardiac surgery. Differential glucose consumption protein cargo fat-released exosomes, after semaglutide or/and insulin treatment analyzed by enzymatic TripleTOF, respectively. Human phenotype their adhesion (HAEC) or angiogenesis flow cytometry functional fluorescence analysis. Immune plasma markers determined Luminex-multiplex before 6 months semaglutide. Results GLP-1 was expressed neutrophils. exosomes-protein identified explants treatment. This drug secretion gelsolin, antithrombotic protein, EAT, modulated CD11b neutrophils, migration adhesion, induced adiposity FABP4, chemoattractant. Monocytes adiposity, stretch, mesothelial, fibrotic, inflammatory underwent for showed 20% reduction statistical significance FABP4 levels 80% increase neutrophils-CD88. Conclusion Semaglutide increases endocrine activity properties. Moreover, this modulates the atherogenic profile marker, which is also reduced
