Today, air pollution is one of the greatest threats to organismal healthspan. The environmental earth contaminated with a wide variety artificially generated pollutants like fine particulate matter (PM2.5) emitting from industry, fuel engine vehicles, biomass combustion, fumes blasting, crop residue burning, and wildfire. pollutant PM2.5 induces massive oxidative stress inflammation, major contributors in initiation progression numerous diseases including pulmonary, cardiovascular, renal, hepatic, reproductive, neurological, mental, accelerated biological aging. provocative question following: how can we solve this associated problem? As it not realistic clean environment at once toxic pollution, initiatives have been undertaken develop novel therapeutic approaches control air-pollutant-induced inflammation devastating diseases. primary goal review article discuss systematically key findings recent preclinical studies documenting first, role augmentation stress, diseases; second, efficacies different natural synthetic compounds amelioration PM2.5-induced pyroptosis, pathologies. Further investigation on safety these will be helpful select effective non-toxic compound(s) for clinical trial drug development.

Fine particulate matter (PM2.5)-related health issues have received increasing attention as a worldwide public problem, and PM2.5-related chronic kidney disease (CKD) has been emerging over the years. Limited research focused on mechanism of PM2.5-induced disease. To investigate impact PM2.5 its potential mechanism, we generated PM2.5-exposed C57BL/6 mouse model by using Shanghai Meteorological Environment Animal Exposure System (Shanghai-METAS) for 12 weeks, urine, blood tissues were collected. The pathological changes function measured after exposure weeks. Along with glomerular damage, tubular damage was also severe in mice. results mRNA-seq indicate that pyroptosis is involved. Pyroptosis defined caspase-1-dependent programmed cell death response to insults. expression nucleotide-binding oligomerization domain-like receptor family pyrin domain containing 3 (NLRP3), Caspase-1, gasdermin D (GSDMD) IL-1β detected. NLRP3 inflammasome activation subsequent observed Bumpt cells too. At meantime, inhibitors caspase-1 applied exposed cells. It turned out significant rescue effect inhibitors. This study revealed new insights into injury specific well morphological changes, important role dysfunction.

Eleutheroside E (EE) is a primary active component of Acanthopanax senticosus, which has been reported to inhibit the expression inflammatory genes, but underlying mechanisms remain elusive. High-altitude pulmonary edema (HAPE) severe complication high-altitude exposure occurring after ascent above 2500 m. However, effective and safe preventative measures for HAPE still need be improved. This study aimed elucidate potential mechanism EE in HAPE. Rat models were established through hypobaric hypoxia. Mechanistically, hypoxia aggravates oxidative stress upregulates (pro)-inflammatory cytokines, activating NOD-like receptor protein 3 (NLRP3) inflammasome-mediated pyroptosis, eventually leading suppressed NLRP3 pyroptosis by inhibiting nuclear translocation factor kappa-Β (NF-κB), thereby protecting lung from nigericin (Nig), an activator, partially abolished protective effects EE. These findings suggest promising agent preventing induced pyroptosis.

Asthma is a common chronic respiratory disease, and its treatment core problem challenge in clinical practice. Glucocorticoids (GCs) are the first-line therapy for of asthma. Local systemic adverse reactions caused by GCs create obstacles to Therefore, research target find new, safe, effective therapeutic medicine at present. Natural products an important source treating asthma with low cost toxicity. Astragaloside IV (AS-IV) active ingredient traditional Chinese Astragalus mongholicus Bunge. Previous studies have indicated that AS-IV plays role inhibiting airway inflammation remodeling airway, regulating immunity neuroendocrine function (Fig. 1) . It has variety biological characteristics such as multi-target intervention, high safety, good curative effect. This article reviews specific mechanism provide references subsequent research.

Air pollution has been broadly concerned as a global issue in all regions. Particulate matter (PM) emission mainly originated from the combustion of diesel engine transport sector is progressively solved for mitigation. The purpose this study to preliminary utilize biodiesel different feedstock an alternative fuel engine. Waste-derived was prepared waste cooking oil (WCOME) and chicken (CKOME) by transesterification process. Palm oil-derived (POME) also used reference. Fatty acid profiles were characterized gas chromatography–mass spectrometer. analyzed characteristics under ASTM standards. test carried out on single-cylinder at speed 1500 rpm with full load. performance well exhaust emissions waste-derived compared palm fuel. PM2.5 adversely human health through mechanisms such as oxidative stress, inflammatory response, autophagy, ferroptosis, endoplasmic reticulum stress. Phytochemicals are of interest for their broad range physiological activities few side effects, and, recent years, they been widely used mitigate the adverse effects caused by exposure. In this review, roles phytochemicals summarized, including those polyphenols, carotenoids, organic sulfur compounds, saponin mitigating PM2.5-induced reactions different molecular mechanisms, anti-inflammatory antioxidant inhibition stress regulation autophagy. These useful a scientific basis prevention treatment disease PM2.5.

Long-term exposure to noise can cause irreversible hearing loss. Considering that there is no effective drug treatment, it important seek preventive treatment for noise-induced loss (NIHL). Although astragaloside IV (AS-IV) protects against NIHL by reducing serum inflammatory factors, scarce information on the regulation of factors AS-IV prevent NIHL. We investigated thresholds and relationship between levels cytokines intestinal microbiota c57bl/6j mice exposed (103 dB SPL 4 h·d−1) 7 days, treated with or without AS-IV. Our results revealed a lower threshold TNF-α, TNF-γ, IL-6, IL-1β, IFN-γ in Additionally, increased abundance phylum Firmicutes, class Bacillus, order Lactobacillus, family Lactobacillus (p < 0.05), decreased those Bacteroidetes Bacteroidales 0.05). Bacilli negatively correlated IL-1β; Erysipelotrichaceae INF-γ; Clostridiales positively IL-1β. In conclusion, reduces elevation mice, preventing noise, under intervention AS-IV, changes correlate flora. suggest improves flora inflammation noise.
