Abstract Ferroptosis, an iron-dependent form of cell death characterized by uncontrolled lipid peroxidation, is governed molecular networks involving diverse molecules and organelles. Since its recognition as a non-apoptotic pathway in 2012, ferroptosis has emerged crucial mechanism numerous physiological pathological contexts, leading to significant therapeutic advancements across wide range diseases. This review summarizes the fundamental mechanisms regulatory pathways underlying ferroptosis, including both GPX4-dependent -independent antioxidant mechanisms. Additionally, we examine involvement various conditions, cancer, neurodegenerative diseases, sepsis, ischemia–reperfusion injury, autoimmune disorders, metabolic disorders. Specifically, explore role response chemotherapy, radiotherapy, immunotherapy, nanotherapy, targeted therapy. Furthermore, discuss pharmacological strategies for modulating potential biomarkers monitoring this process. Lastly, elucidate interplay between other forms regulated death. Such insights hold promise advancing our understanding context human health disease.

Dual-state emissive (DSE) materials exhibit fluorescence in both solid and solution states have become an emerging material the fields of science sensing recent years. However, due to lack effective universal preparation methods, DSE materials, especially those with long emission wavelengths, are still scarce. Developing method for constructing such molecules is urgently needed. In this study, we constructed three (NRP-Boc, DCIP-Boc, DCMP-Boc) far-red near-infrared by simply modifying traditional aggregation-caused quenching (ACQ) fluorophores tert-butyloxycarbonyl (Boc) groups. Density functional theory (DFT) calculations crystal data revealed reasons bright these solid, demonstrating that Boc protection a simple strategy molecules. Myocardial ischemia/reperfusion injury (MIRI) is the leading cause of irreversible myocardial damage. A pivotal pathogenic factor (I/R)-induced cardiomyocyte ferroptosis, marked by iron overload and lipid peroxidation. However, impact droplet (LD) changes on I/R-induced ferroptosis unclear. In this study, an aggregation-induced emission probe, TPABTBP developed that used for imaging dynamic in LD during ferroptosis. exhibits excellent LD-specificity, superior capability monitoring lipophagy, remarkable photostability. Molecular dynamics (MD) simulation super-resolution fluorescence demonstrate specifically localized to phospholipid monolayer membrane LDs. Imaging LDs cardiomyocytes tissue model mice with MIRI reveals accumulation level increase early reperfusion stage (0-9 h) but decrease late (>24 via lipophagy. The inhibition breakdown significantly reduces peroxidation cardiomyocytes. Furthermore, it demonstrated chloroquine (CQ), FDA-approved autophagy modulator, can inhibit thereby attenuating mice. This study describes ischemia suggests a potential therapeutic target intervention.
