There exists a bidirectional relationship between oral health and general well-being, with an imbalance in symbiotic flora posing threat to overall human health. Disruptions the commensal can lead diseases, while systemic illnesses also impact cavity, resulting development of diseases disorders. Porphyromonas gingivalis Fusobacterium nucleatum , known as pathogenic bacteria associated periodontitis, play crucial role linking periodontitis accompanying diseases. In periodontal tissues, these bacteria, along their virulence factors, excessively activate host immune system through local diffusion, lymphatic circulation, blood transmission. This response disruption contributes osteoimmune mechanisms, alveolar bone resorption, potential inflammation. To restore homeostasis, deeper understanding microbiota–host interactions network phenotype tissues is imperative. Defining offers promising avenue for investigating complex characteristics plaque biofilms exploring review aims provide overview mechanisms underlying - -induced well immunophenotypes observed during pathological conditions.

In patients with rheumatoid arthritis (RA), intestinal flora imbalance and butyrate metabolism disorders precede clinical are associated the pathogenesis of RA. This can alter immunology permeability mucosa, leading to damage barrier. this context, bacteria their metabolites enter bloodstream reach distant target tissues host, resulting in local inflammation aggravating arthritis. Additionally, is also exacerbated by bone destruction immune tolerance due disturbed differentiation osteoclasts adaptive cells. Of note, a metabolite flora, which not only locally inhibits immunity targets zonulin tight junction proteins alleviate barrier-mediated but autoantibodies balances responses T B lymphocytes throughout body repress erosion inflammation. Therefore, key intermediate linking host. As result, restoring butyrate-producing capacity using exogenous potential therapeutic strategies for RA future.

Mesenchymal stem cell exosome (MSCs-exo) is a class of products secreted by mesenchymal cells (MSCs) that contain various biologically active substances. MSCs-exo promising alternative to MSCs due their lower immunogenicity and lack ethical constraints. Ginsenoside Rh2 (Rh2) hydrolyzed component the primary substance ginsenosides. has variety pharmacological functions, including anti-inflammatory, anti-tumor, antioxidant. Studies have demonstrated gut microbiota metabolites are critical in developing rheumatoid arthritis (RA). In this study, we constructed collagen-induced (CIA) model rats. We used combined with treat CIA To observe effect on joint inflammation, rat feces were collected for 16 rRNA amplicon sequencing untargeted metabolomics analysis. Increased abundance of Segatella copri (S. copri) within the gut microbiota is associated with systemic inflammatory diseases, including rheumatoid arthritis. Although outer-membrane vesicles (OMVs) Gram-negative bacteria are important players in microbiota–host communication, effect S. copri-derived OMVs on immune cells unknown. Macrophages engulf and eliminate foreign material conditioned by environmental signals to promote either homeostasis or inflammation. Thus, we aimed explore impact copri-OMVs human macrophages vitro, employing THP-1 monocyte-derived macrophage models. The uptake DiO-labeled into was monitored confocal microscopy flow cytometry. Furthermore, phenotype cytokine secretion naïve (M0), pro-inflammatory (M1), anti-inflammatory (M2) analyzed cytometry ELISA, respectively. We show that enter through macropinocytosis clathrin-dependent mechanisms. copri-OMVs, but not parental bacterium, induced a dose-dependent increase expression M1-related surface markers M0 M2 activated large amounts cytokines M1 macrophages. These results highlight an role promoting responses, which might contribute diseases.

The increase of the Fusobacterium nucleatum level has been previously identified in various cancers including gastric cancer (GC), but how F. exerts its carcinogenic role GC remains unclear. Several studies revealed that contributes to progression via secretion extracellular vehicles (EVs). Hence, it's designed reveal influence nucleatum-derived EVs (Fn-EVs) progression. tumor and adjacent tissues were collected from 30 patients, abundance was found be highly expressed samples. ultracentrifugation employed isolate Escherischia coli (E. coli), which labeled Fn-EVs E. coli-EVs, respectively. After treating cells with cell counting kit 8, colony formation, wound healing as well transwell assay performed, effectively enhanced oxaliplatin resistance, facilitated proliferation, migration, invasion, stemness while coli-EVs exert no significant effect on cells. Besides, DNA repair also by Fn-EVs, sphere-forming detection stemness- repair-associated proteins western blotting. In vivo analyses demonstrated administration not only promoted growth liver metastasis conferred resistance resistance. This study first contributive development provided a better perspective for manipulating patients malignant phenotypes.
