Gut microbiota includes a vast collection of microorganisms residing within the gastrointestinal tract. It is broadly recognized that gut and brain are in constant bidirectional communication, which its metabolic production major component, form so-called microbiome–brain axis. Disturbances homeostasis caused by imbalance their functional composition activities, known as dysbiosis, cause dysregulation these pathways trigger changes blood–brain barrier permeability, thereby causing pathological malfunctions, including neurological disorders. In turn, can affect structure function through autonomic nervous system regulating motility, intestinal transit secretion, permeability. Here, we examine data from CAS Content Collection, largest published scientific information, analyze publication landscape recent research. We review advances knowledge related to human microbiome, complexity functionality, communication with central system, effect axis on mental health. discuss correlations between various diseases, specifically also explore metabolites regard impact associated diseases. Finally, assess clinical applications gut-microbiota-related substances development pipelines. hope this serve useful resource understanding current emerging field an effort further solving remaining challenges fulfilling potential.

Intestinal dysbiosis frequently occurs in abdominal radiotherapy and contributes to irradiation (IR)-induced intestinal damage inflammation. Akkermansia muciniphila (A. muciniphila) is a recently characterized probiotic, which critical for maintaining the dynamics of mucus layer preserving microbiota homeostasis. However, role A. alleviation radiation enteritis remains unknown. In this study, we reported that abundance was markedly reduced intestines mice exposed IR feces patients who received radiotherapy. Abundance negatively correlated with duration diarrhea patients. Administration substantially mitigated IR-induced prevented mouse death. Analyzing metabolic products revealed propionic acid, short-chain fatty acid secreted by microbe, mediated radioprotective effect. We further demonstrated bound G-protein coupled receptor 43 (GRP43) on surface epithelia increased histone acetylation hence enhanced expression tight junction proteins occludin ZO-1 elevated level mucins, leading integrity epithelial barrier radiation-induced damage. Metformin, first-line agent treatment type II diabetes, promoted through increasing muciniphila. Together, our results plays reduction Application probiotics or their regulators, such as metformin, could be an effective protection exposure-damaged intestine.

The human gastrointestinal tract hosts a complex and dynamic community of microorganisms known as the gut microbiota, which play pivotal role in numerous physiological processes, including digestion, metabolism, immune function. Recent research has highlighted significant impact diet on microbiota composition functionality, consequential effects host health. Concurrently, there is growing evidence linking to inflammation, key factor many chronic diseases such inflammatory bowel disease (IBD), obesity, diabetes, cardiovascular (CVDs). This review explores how dietary components influence composition, these microbial changes affect pathways, therapeutic implications modulating this axis for prevention management. Beneficial patterns, Mediterranean (MD) plant-based diets, promote diverse balanced supporting anti-inflammatory pathways. Conversely, Western (WD), high saturated fats refined sugars, associated with dysbiosis increased inflammation. With all links between three variables considered, attempts offer thorough examination triangle formed by food.

Despite significant progress in cancer treatment, traditional therapies still face considerable challenges, including poor targeting, severe toxic side effects, and the development of resistance. Recent advances biotechnology have revealed potential bacteria their derivatives as drug delivery systems for tumor therapy by leveraging biological properties. Engineered bacteria, Escherichia coli, Salmonella, Listeria monocytogenes, along with derivatives─outer membrane vesicles (OMVs), bacterial ghosts (BGs), spores (BSPs)─can be loaded a variety antitumor agents, enabling precise targeting sustained release within microenvironment (TME). These possess intrinsic properties that stimulate immune system, enhancing both innate adaptive responses to further amplify therapeutic effects. The ability naturally accumulate hypoxic regions versatility genetic modifications allow tailored strategies synergistically enhance effectiveness chemotherapy, immunotherapy, targeted therapies. This review comprehensively examines fundamental principles therapy, focusing on employed engineering, loading, use therapy. It also discusses challenges faced optimizing systems, such safety concerns, unintended responses, scalability clinical applications. By exploring these aspects, this provides theoretical framework improving bacterial-based contributing more effective personalized treatments.

Akkermansia muciniphila is a mucin-degrading bacterium of the intestinal niche, exerting beneficial effects on host metabolic profile. Accumulating evidence indicated as promising therapeutic probiotic against disorders such obesity, type 2 diabetes and cardiovascular diseases. However, in specific microenvironments, its excessive enrichment may be not beneficial. Conditions like inflammatory bowel disease (IBD), Salmonella typhimurium infection or post-antibiotic reconstitution benefit from supplementation. Furthermore, using patients with endocrine gynecological disorders-such polycystic ovary syndrome (PCOS) endometriosis-that have higher risk developing IBD, should critically evaluated. In addition, cautionary note comes neurological field, gut microbiota suffering Parkinson's multiple sclerosis exhibits characteristic signature municiphila abundance. Overall, considering these controversial points, use evaluated an individual basis, avoiding risking unexpected effects.

Increasing evidence suggests that the gut microbiota may represent potential strategies for Parkinson’s disease (PD) treatment. Our previous research revealed a decreased abundance of Akkermansia muciniphila (Akk) in PD mice; however, whether Akk is beneficial to unknown. To answer this question, mice received MPTP intraperitoneally construct subacute model and were then supplemented with orally 21 consecutive days. Motor function, dopaminergic neurons, neuroinflammation, neurogenesis examined. In addition, intestinal inflammation, serum fecal short-chain fatty acids (SCFAs) analyses, assessed. We found treatment effectively inhibited reduction neurons substantia nigra pars compacta (SNpc) partially improved motor function mice. Additionally, markedly alleviated neuroinflammation striatum hippocampus promoted hippocampal neurogenesis. It also level colon inflammation. Furthermore, these aforementioned changes are mainly accompanied by alterations isovaleric acid levels, lower permeability. strongly neuroprotective agent therapy.
