The maintenance of epithelial barrier function involves cellular tension, with cells pulling on their neighbors to maintain integrity. Wounding interrupts which may serve as an early signal initiate repair. To characterize how wounds alter we used a laser-recoil assay map cortical tension around in the monolayer Drosophila pupal notum. Within minute wounding, there was widespread loss along both radial and tangential directions. This similar levels observed Rok inactivation. Tension subsequently restored wound, first distal then proximal cells, reaching wound margin about 10 minutes after wounding. Restoring required GPCR Mthl10 IP3 receptor, indicating importance this calcium signaling pathway known be activated by damage. restoration correlated inward-moving contractile wave that has been previously reported; however, itself not affected knockdown. These results indicate transiently increase contract absence signaling, but is critical for fully resetting baseline it disrupted

Basement membranes are sheets of extracellular matrix separating tissue layers and providing mechanical support. Their properties determined largely by their most abundant protein, Collagen IV (Col4). Although basement repaired after damage, little is known about how. To wit, since membrane it unknown how damage detected, Col4 long-lived regulated to avoid fibrosis. Using the adult

The pupae of Drosophila melanogaster are immobile for several days during metamorphosis, which they develop a new body with thin transparent adult integument. Their immobility and transparency make them ideal in vivo live imaging experiments. Many studies have focused on the dorsal epithelial monolayer pupal notum because its accessibility relatively large size. In addition to mechanics development, has been an tissue study wound healing. After injury, entire repair process can be captured by over 6-12 h. Despite popularity imaging, very few published utilized fixed samples. Fixation staining common approaches nearly all other tissues, taking advantage repertoire simple cellular stains antibodies. However, is fragile prone curling distortion after removal from body, making it challenging complement imaging. This protocol offers straightforward method fixing notum, both intact laser-wounding. With this technique, ventral side pupa glued down coverslip immobilize pupa, carefully removed, fixed, stained. epithelium mounted slide or between two coverslips facilitate tissue's side.
