Per- and polyfluoroalkyl substances (PFASs) have been found in toilet paper Europe, the United States, Africa; however, their presence Chinese household has not investigated. In this study, 21 legacy 30 emerging PFASs were analyzed tissue from China, including 48 samples of different origins materials. Median concentrations chlorinated polyfluorinated ether sulfonates (Cl-PFESAs), hexafluoropropylene oxide homologs (HFPOs), p-perfluorous nonenoxybenzenesulfonate (OBS) 0.32, 0.19, 0.10 ng/g dry weight (dw), respectively. The detection frequencies (DFs) Cl-PFESAs OBS both greater than 96%, followed by trimer acid (HFPO-TrA, DF: 85%). Notably, level HFPO-TrA one sample was as high 540 dw, indicating a potential environmental risk to humans. addition, significantly higher that (p < 0.05). However, no significant differences observed materials origins. This suggests PFASs, particularly are widely detected products across is source landfill contamination human exposure.

The biotoxicity of perfluoroalkyl and polyfluoroalkyl substances (PFASs) to aquatic organisms has been widely concerned. However, studies on toxic effects PFASs are usually evaluated directly by using laboratory exposure rather than validation based data obtained in the field. In this study, wild catfish (Silurus meridinalis) was explored relationship between bioaccumulation lipid disorders. Nine thirteen metabolites were significantly associated with perfluorooctane sulfonate (PFOS) 6:2/8:2Cl-PFESA (trade name F-53B) exposures, respectively; correlated fatty acid (FA) conjugates, FA esters, steroids, glycerophosphate subclasses. metabolism fish its mechanism further analyzed through experiments. Zebrafish (Danio rerio) different sexes underwent PFOS F-53B exposures for 21 days at 100 ng/L μg/L. By determining gene expression levels, hepatic contents, histopathological change, adverse order male or female μg/L/F-53(−|-)B > μg/L ng/L/F-53(−|-)B PFOS; stress response more intensive that female. activated peroxisome proliferator-activated receptor pathway, promoting processes total cholesterol (T-CHO) transport, β-oxidation, synthesis, finally induced T-CHO transportation from blood into liver, then accelerated ester transformation, CHO steroids. Laboratory experiments confirmed field analysis. This study innovatively their mechanisms highlighting concerns regarding health risks.

Globally implemented ecological risk assessment (ERA) guidelines marginalize hormesis, a biphasic dose–response relationship characterized by low-dose stimulation and high-dose inhibition. The present study illuminated the promise of hormesis as scientific model for ERA per- polyfluoroalkyl substances (PFAS) represented perfluorooctanoic acid (PFOA) perfluorooctanesulfonic (PFOS). A total 266 hormetic relationships were recompiled from 1237 observations, covering 30 species nine representative taxonomic groups. standardized amplitudes followed log-normal probability distribution, being subject to limits biological plasticity but independent stress inducers. SHapley Additive exPlanations algorithm revealed that target endpoint was most important variable explaining amplitudes. Subsequently, quantitative frameworks established incorporate into predicted no-effect concentration levels, with lower induction dose zero-equivalent point broader zone PFOS. Realistically, 10,117 observed concentrations PFOA PFOS gathered worldwide, 4% which fell within zones, highlighting environmental relevance hormesis. Additionally, potential identified in other legacy emerging PFAS well their alternatives mixtures. Collectively, it is time concept studies facilitate more realistic characterizations.

Perfluorohexane sulfonic acid (PFHxS) is extensively used in waterproof coatings and fire-fighting foams, several studies have found it to be a potential health hazard, but there still unknown about its effects on spermatogenesis. Our results showed that PFHxS-treated mice significant reproductive toxicity, including decrease sperm count motility, the levels of sex hormones (P < 0.05). Concurrently, structural abnormalities are observed sperm, affecting ≈60-75% those group. Additionally, structure blood-testis barrier (BTB) damaged after PFHxS treatment, leading higher expression inflammatory cytokines microenvironment for Moreover, proteins associated with mitochondrial biogenesis, PTEN-induced kinase 1 (PINK1) NADPH oxidase 4 (NOX4), dysregulated testes treatment. Based metabolome data, differential metabolite 3-hydroxybutanoic identified group, which can regulate histone Kac levels, especially H3K4ac H3K9ac. In summary, this study suggest mice, factors disrupt function metabolic profiles hinder progress gene transcription through Kac, ultimately causing dysfunction.

Per- and polyfluoroalkyl substances (PFAS) pose potential health risks to lipid metabolism, but the effects of emerging PFAS alternatives, particularly in children, remain unclear. This cross-sectional study investigated association between exposure levels 294 Chinese children aged 7-10 years, analyzing blood samples for 14 profiles, including triglycerides (TG), total cholesterol (TC), high-density lipoprotein (HDL), low-density (LDL), apolipoprotein A1 (ApoA1), B (ApoB). Exposure 6:2 Cl-PFESA, PFO4DA, PFO5DoDA was associated with higher TC, TG, LDL levels, PFO4DA increasing TC by 1.7% TG 10.7%. Weighted quantile sum (WQS) regression showed mixed positively (0.08, 95% CI: 0.007, 0.153). had highest weight (0.468), (0.327), (0.57), ApoA1 (0.243), ApoB (0.466), while PFMOAA HDL (0.332). Bayesian Kernel Machine Regression (BKMR) analysis confirmed positive associations mixture LDL, ApoA1. Mediation revealed that mtDNAcn significantly mediated exposure's effect on explaining 27.2-74.2% effect. These findings highlight need regulatory action address risks.
