Muscle wasting occurs with aging and may be a result of oxidative stress damage potentially inadequate protection by lipophilic antioxidants, such as vitamin E. Previous studies have shown muscular abnormalities behavioral defects in E-deficient adult zebrafish. To test the hypothesis that there is an interaction between muscle degeneration caused E deficiency, we evaluated long-term deficiency skeletal zebrafish using metabolomics. Zebrafish (55 days old) were fed E+ E− diets for 12 or 18 months. Then, samples analyzed UPLC-MS/MS. Data to highlight metabolite pathway changes seen either status both. We found altered purines, various amino acids, DHA-containing phospholipids. Vitamin at months was associated acid metabolism, specifically tryptophan pathways, systemic regulation purine In sum, while both induced did some overlap dysregulated metabolic each factor also presented unique alterations, which require further study more confirmatory approaches.

Abstract The unprecedented rise in global ambient temperatures the last decade has significantly impacted human health, yet how heat exposure affects development of sarcopenia remains enigmatic. Here, we demonstrate that chronic induces skeletal muscle volume loss, leading to strength and functional decline mice. microbiota composition heat‐exposed mice was analyzed using 16S ribosomal DNA analysis. Liquid chromatography‐mass spectrometry (LC–MS) used explore effects on blood metabolome further analyze correlation between metabolism gut microbiota. Transplantation from germ‐free sufficient increase adverse function host. Mechanistically, an untargeted metabolomics strategy, reveal altered due high is associated with elevated serum levels homocitrulline. Homocitrulline causes mitochondrial dysfunction myocytes by exacerbating ferroptosis levels. And Nrf2 activator (Oltipraz) supplementation alleviates atrophy induced exposure. Our findings detrimental provide new strategies for treating sarcopenia.

The significance of complex I the electron transport chain (ETC) in aging process is widely acknowledged; however, its specific impact on development sarcopenia muscle remains poorly understood. This study elucidated correlation between inhibition and by conducting a comparative analysis skeletal gene expression phenotypes from rats, mice, humans. Our findings reveal common mechanistic link across species, particularly highlighting suppression ETC activity dysregulated mitochondrial transcription translation phenotypes. Additionally, we observed macrophage dysfunction alongside abnormal metabolic processes within tissues all implicating their pathogenic role onset sarcopenia. These discoveries underscore importance understanding shared mechanisms associated with development. identified correlations provide valuable insights into potential targets for therapeutic interventions aimed at mitigating sarcopenia, condition substantial implications populations.

Sarcopenia is one of the primary risk factors for various adverse health events in later life. However, its pathophysiology very old population remains unclear. Hence, this study aimed to examine whether plasma free amino acids (PFAAs) correlate with major sarcopenic phenotypes (i.e., muscle mass, strength, and physical performance) community-dwelling adults aged 85-89 years living Japan. Cross-sectional data from Kawasaki Aging Well-being Project were used. We included 133 years. In study, fasting blood was collected measure 20 PFAAs. Measures three appendicular lean mass assessed by multifrequency bioimpedance, isometric handgrip gait speed a 5 m walk at usual pace. Furthermore, we used phenotype-specific elastic net regression models adjusted age centered 85 years, sex, body index, education level, smoking status, drinking habit identify significant PFAAs each phenotype. Higher histidine lower alanine levels associated poor speed, but no correlated strength or mass. conclusion, such as are novel biomarkers performance older.

Abstract Background Various biomarkers associated with sarcopenia have been identified. However, there is a scarcity of studies exploring and validating in individuals age-related sarcopenia. Aims This study aimed to investigate the proteome identify potential for Methods Proteomic analysis experimental validation were conducted using plasma from hospitalized older adults. Sarcopenia diagnosis was based on Asian Working Group 2019 criteria. Data-independent acquisition-based proteomics performed 60 participants, 30 diagnosed without Differentially expressed proteins (DEPs) selected evaluated by Receiver Operating Characteristic (ROC) analysis. Biomarker candidates further quantitatively validated enzyme-linked immunosorbent assay (ELISA) utilizing 6 participants Results A total 39 DEPs identified 12 ROC 8 included ELISA their predictive performance. Paraoxonase-3 (PON3) consistently showed down-regulation sarcopenic group across both methodologies. Insulin-like growth factor-binding protein-2 (IGFBP2) inconsistency group, up-regulation observed proteomic but ELISA. Discussion Decline PON3 may result an overload oxidative stress skeletal muscles contribute Protein modifications IGFBP2 might exhibit during pathogenesis. Conclusions Plasma are implicated highlighted as biomarker patients Further imperative gain in-depth understanding IGFBP2.
