The potential biological function of tea and its active components on colitis has attracted wide attention. In this study, different ingredients including polyphenols (TPPs), polysaccharides (TPSs), theabrownin (TB), theanine (TA) have been compared in the intervention dextran sulfate sodium (DSS)-induced mice. Specifically, TPP showed greatest effect since it reduced 60.87% disease activity index (DAI) to that DSS-induced group, followed by reduction 39.13% TPS 28.26% TB DAI, whereas there was no obvious alleviative TA colitis. The stromal interaction molecules (STIMs) are the sarcoplasmic reticulum (SR) Ca2+ sensors that trigger store-operated entry (SOCE) in a variety of cell types. While STIM1 isoform has been focus research cardiac pathophysiology, function homolog STIM2 remains unknown. Using imaging and patch-clamp techniques, we showed knockdown (KD) by siRNAs increased SOCE ISOC current neonatal rat ventricular cardiomyocytes (NRVMs). Within this cardiomyocyte model, identified transcript expression Stim2.1 Stim2.2 splice variants, with predominance for Stim2.2. conventional super-resolution confocal microscopy (STED), found exogenous STIM2.1 STIM2.2 formed pre-clusters reticular organization at rest. Following SR store depletion, some clusters were translocated to SR-plasma membrane (PM) junctions co-localized Orai1. overexpression strategy revealed suppressed Orai1-mediated while enhanced SOCE.
