Abstract BackgroundThe senescence of mesenchymal stem cells (MSCs) greatly compromises their therapeutic effect in regenerative medicine. Numerous studies are focusing on possible rejuvenation strategies to enhance the efficacy autologous MSC-based therapy. S-adenosyl-L-methionine (SAM) is a metabolite present all living cells. However, there little research about effects and mechanisms SAM MSCs.MethodsIn this study, adipose-derived MSC (ADSC) were assessed vitro by β-galactosidase staining, reactive oxygen species assay, cell cycle tests, Western blot. The osteogenic adipogenic differentiation ability changes also detected. Mouse models premature aging established subcutaneous injection D-Galactose (D-gal). vivo antiaging roles tested through behavioral organ coefficient, pathological morphology, expression aging-related proteins major organs, such as heart, liver, kidney. bone density distal femur, well volume, number, thickness trabecula evaluated micro-CT. molecular searched validated transcriptome sequencing, blot, immunofluorescence. In addition, level ROS promoted ADSCs. Moreover, D-gal-induced mouse model aging, exercise kidney mice. Additionally, number trabeculae femur mice increased. Transcriptome sequencing results revealed that PI3K/AKT involved SAM-mediated anti-senescence MSCs. Mechanistically, activated signaling increased phosphorylation FOXO3a, resulting decrease translocation FOXO3a nucleus inhibition activity P21. Ultimately, attenuated ADSCs potential.ConclusionsIn summary, our suggested could ameliorate MSCs PI3K/AKT/FOXO3a signaling. Meanwhile, exert vivo.

Previous reports have confirmed that saponins (ginsenosides) derived from Panax ginseng. C. A. Meyer exerted obvious memory-enhancing and antiaging effects, the simpler structure of ginsenosides, better biological activity. In this work, we aimed to explore therapeutic effect underlying molecular mechanism 20(S)-protopanaxatriol (PPT), aglycone panaxatriol-type by establishing D-galactose (D-gal)-induced subacute brain aging model in mice. The results showed PPT treatment (10 20 mg/kg) for 4 weeks could significantly restore D-gal (800 mg/kg 8 weeks)-induced impaired memory function, choline dysfunction, redox system imbalance Meanwhile, also reduced histopathological changes caused exposure. Moreover, increase TFEB/LAMP2 protein expression promote mitochondrial autophagic flow. Importantly, docking had good binding ability with LAMP2 TFEB, suggesting might play an important role alleviate D-gal-caused aging.

Heat stress is a major problem that constrains pig productivity. Understanding and identifying adaptation to heat has been the focus of recent studies, identification genome-wide selection signatures can provide insights into mechanisms environmental adaptation. Here, we generated whole genome re-sequencing data from six Chinese indigenous populations identify genomic regions with related tolerance using multiple methods: three methods for intra-population analyses (Integrated Haplotype Score, Runs Homozygosity Nucleotide diversity Analysis) (iHS, ROH, Pi) inter-population (Fixation index, Cross-population Composite Likelihood Ratio Extended Homozygosity) (FST, XP-CLR, XPEHH). In total, 1 966 796 single nucleotide polymorphisms (SNPs) were identified in this study. Genetic structure FST indicated differentiation among these breeds. Based on information location environment, breeds divided cold groups. By combining two or more approaches signatures, outlier signals overlapping as candidate regions. A total 163 genes identified, which, 29 associated injury anti-inflammatory effects. These further 78 Gene Ontology (GO) functional terms 30 Kyoto Encyclopedia Genes Genomes (KEGG) pathways enrichment analysis (P < 0.05). Radiation-induced lung injury (RILI) is a common side effect in thoracic tumor patients undergoing radiotherapy. At present, there no ideal radio-protective agent which widely used RILI treatment. Astilbin (AST), bioactive flavonoid, exhibits various biological effects, including anti-inflammatory, antioxidant, and anti-fibrotic activities, partly result from reducing oxidative stress inflammation pathogenic conditions. However, the protective efficacy of AST to ameliorate has not been reported. In this study, we employed network pharmacology, RNA sequencing, experimental evaluation reveal effects pharmacological mechanism treat vivo vitro. We observed that reduced radiation-induced apoptosis, DNA damage, inflammatory reactions, reactive oxygen species (ROS) level human normal epithelial cells BEAS-2B. Further study showed treatment significantly ameliorated by pathology changes reaction tissue C57BL/6J mice. Mechanistically, expression epithelial-mesenchymal transition (EMT) markers radiation-triggered acetylation p53 protein were alleviated Furthermore, after intervention Trichostatin A (TSA). Our data indicate can alleviate inhibiting reactions EMT process through decreasing acetylation. conclusion, our suggests great potential be new therapeutic compound for RILI.

Objective To investigate the effects of phillyrin on D-galactose (D-gal)-induced aging in brain mice and mechanism. Methods Institute Cancer Research were intraperitoneally injected with D-gal 150 mg/kg to induce model, treated 5, 15, 45 once a day for 8 weeks. Results The memory impairment induced by was ameliorated treatment, oxidative stress serum inhibited, as evidenced increased superoxide dismutase, glutathione peroxidase enzyme activities, decreased malondialdehyde levels. Additionally, hippocampus damage has also been well improved phillyrin. Moreover, protein expressions nuclear factor erythroid-2-related 2 (Nrf2), heme oxygenase-1 (HO-1), NADPH quinone oxidoreductase 1 (NQO1) markedly up-regulated expression Keap-1 down-regulated group than that model group. Conclusion These findings suggest is effective prevention age-related impairment, attenuation activating Nrf2/HO-1 signaling pathway may be one mechanisms.
