ABSTRACT Introduction COVID-19 is a rapidly spreading infectious disease caused by the SARS-CoV-2 virus. Although several therapeutic interventions have been developed, mortality rate of remains high, and effective treatment options are urgently needed. Angiotensin-converting enzyme 2 receptors (ACE2R) are requisite to enter the host cells for severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). ACE2R is constitutive and functions as a type I transmembrane metallo-carboxypeptidase in renin-angiotensin system (RAS). On thyroid follicular cells, allows SARS-CoV-2 invade gland, impose cytopathic effects produce endocrine abnormalities, including stiff back, neck pain, muscle ache, lethargy, enlarged, inflamed gland COVID-19 patients. Further damage perpetuated by sudden bursts of pro-inflammatory cytokines, which suggestive life-threatening known "cytokine storm". IL-1β, IL-6, IFN-γ, TNF-α identified key orchestrators cytokine storm. These inflammatory mediators upregulate transcriptional turnover nuclear factor-kappa B (NF-κB), Janus kinase/signal transducer activator transcription (JAK/STAT), mitogen-activated protein kinase (MAPK), paving pathway storm-induced dysfunctions euthyroid sick syndrome, autoimmune diseases, thyrotoxicosis Targeted therapies with corticosteroids (dexamethasone), JAK inhibitor (baricitinib), nucleotide analogue (remdesivir) N-acetyl-cysteine have demonstrated effectiveness terms attenuating severity frequency dysfunctions, morbidity mortality Here, we review pathogenesis storms mechanisms pathways that establish connection between disorder COVID-19. Moreover, cross-talk interactions signalling therapeutic strategies address COVID-19-associated diseases also discussed herein.

Inflammatory cytokines and chemokines (CC) drive COVID-19 pathology. Yet, patients with similar circulating CC levels present different disease severity. Here, we determined 171 microRNAomes from 58 hospitalized (Cohort 1) of 25 in the same samples. Combining microRNA (miRNA) measurements allowed for discrimination severe cases greater accuracy than using miRNA or alone. Severity group-specific associations between miRNAs COVID-19-associated (e.g., IL6, CCL20) clinical hallmarks neutrophilia, hypoalbuminemia) separated but Analysis an independent cohort 108 a center 2) demonstrated feasibility CC/miRNA profiling leftover hospital blood samples profiles, revealed CCL20, IL10, miR-451a as key correlates fatal COVID-19. These findings highlight that systemic miRNA/CC networks underpin

The COVID-19 pandemic has disproportionately affected individuals with diabetes mellitus (DM), significantly increasing their risk of adverse outcomes. This retrospective study aimed to explore the underlying factors contributing heightened vulnerability DM severe COVID-19. We reviewed medical records patients diagnosed from August 2020 2022 and identified 60 equally divided into two groups. Group A (n = 30) included those an associated infection, while B served as control group without a infection. Inflammatory biomarkers, venous blood glucose levels, other parameters were assessed at hospital admission again after 14-day treatment period. Statistical analysis confirmed strong association between In treated Empagliflozin, correlations observed IL-1, TNF-alpha, IL-6, levels. Patients in did not show significant improvements inflammatory markers or control. contrast, first group, better interleukin levels demonstrated, suggesting higher success rate for that treatment. Our findings indicate Empagliflozin had limited efficacy managing long-term diabetes-related complications, it might offer benefits acute phase illness.

RNA viruses always have been a serious concern for human health by causing several outbreaks, often pandemics. The excessive mortality and deaths associated with the outbreaks caused these were due to induction of pro-inflammatory cytokines leading cytokine storm. Cytokines are important cell-to-cell communication maintain cell homeostasis. Disturbances this homeostasis can lead intricate chain reactions resulting in massive release cytokines. This could severe self-reinforcement feedback processes, which eventually cause systemic harm, multiple organ failure, or death. Multiple inflammation-associated pathways involved production its regulation. Different induce through interplay their viral factors host proteins miRNAs regulating pathways. review will discuss that play an role regulation storm possible therapeutic potential molecules treatment challenges clinical translation.

The COVID-19 pandemic has been a public health issue around the world in last few years. Currently, there is no specific antiviral treatment to fight disease. Thus, it essential highlight possible prognostic predictors that could identify patients with high risk of developing complications. Within this framework, miRNA biomolecules play vital role genetic regulation various genes, principally, those related pathophysiology Here, we review interaction host and viral microRNAs molecular cellular elements potentiate main pulmonary, cardiac, renal, circulatory, neuronal complications patients. miR-26a, miR-29b, miR-21, miR-372, miR-2392, among others, have associated exacerbation inflammatory process, increasing cytokine storm. In addition, increased expression miR-15b, -199a, -491 are prognosis disease, miR-192 miR-323a were identified as clinical mortality admitted intensive care unit. Finally, address miR-29, miR-122, miR-155, miR-200, therapeutic targets. However, more studies required confirm these findings.

Idiopathic pulmonary fibrosis (IPF) is a chronic progressive lung disease with reduced quality of life and earlier mortality, but its pathogenesis key genes are still unclear. In this investigation, bioinformatics was used to deeply analyze the IPF related genes, so as investigate potential molecular provide guidance for clinical treatment. Next-generation sequencing dataset GSE213001 obtained from Gene Expression Omnibus (GEO), differentially expressed (DEGs) were identified between normal control group. Micro RNAs (miRNAs) have been implicated in the regulation of maturation, proliferation, differentiation, and activation immune cells. In this study, we demonstrated that miR-29a antagonizes IFN-γ production at early times post-LSDV infection cattle. was predicted to target upstream regulators, its inhibition resulted enhanced sensitized peripheral blood mononuclear cells (PBMCs). Further, stimulation PBMCs with LSDV antigen exhibited lower levels miR-29a, concomitant a potent cell-mediated response (CMI), characterized by an increase LSDV-specific CD8+ T cell counts IFN-γ, which eventually facilitated virus clearance. addition, few immunocompromised cattle (developed secondary ~ 6 months) failed mount response, were shown maintain higher levels. Furthermore, as compared crossbred cattle, from Rathi (a native Indian breed) animals along IFN-γ. Finally, analysed ≥ 60% decrease expression correlated CMI response. conclusion, is involved antagonizing LSDV-infected may serve novel biomarker for acute phase infection, well predicting functionality more against than
