Abstract Background Inflammatory factors are associated with depression. We seek to investigate the correlation between inflammatory cytokines and prognosis of depression or suicidal ideation behavior at 3 months in patients. Cyclophosphamide has drastically enhanced the expectancy and quality of life cancer patients. However, it is accompanied by diverse neurological complications which are considered a dose-limiting adverse effect. Neurotoxicity caused cyclophosphamide can manifest in numerous manners including anxiety, depression, motor dysfunction cognitive deficits. This review article offers an overview on cyclophosphamide-induced neurotoxicity, providing unified point view possible underlying molecular mechanisms oxidative brain damage, neuroinflammation, apoptotic neuronal cell death as well disruption balance neurotransmitters neurotrophic factors. Besides, this sheds light promising protective agents that have been investigated using preclinical animal models their biological targets protection mechanisms. Despite results experimental models, none these studied clinical trials. Thus, there lack evidence to advocate use any neuroprotective agent setting. Furthermore, evaluated for its effect anticancer activity tumor-bearing animals. Therefore, great necessity adequate well-designed studies evaluation therapeutic values candidates. In this study, we aimed to investigate the impact of various ultrasound durations on structure and bioactivity sweet corncob polysaccharides treated with ultrasound-assisted degradation using hydrogen peroxide ascorbic acid (H2O2-Vc). We subjected treatment for 0, 30, 60, 90 min alongside H2O2-Vc method. then analyzed their chemical composition structure. Additionally, administered these mice type 2 diabetes (T2DM) through gavage at a dosage 200 mg/kg/day. The results indicated significant reduction in molecular weight degraded polysaccharides, while remained relatively stable. However, basic was retained. vivo experiments demonstrated that had positive T2DM, particularly 60-minute (UH-DSCBP-60 min), which effectively controlled blood glucose levels by regulating glycolipid metabolism livers T2DM. Carbon tetrachloride (CCl4) is a classic chemical hepatotoxicant that triggers liver damage through hepatic exacerbation of oxidative stress. Geraniol (GRL) natural bioactive acyclic monoterpene with several pharmacological effects. We thus explored whether GRL could prevent CCl4-triggered toxicity. Rats were divided and administered (100 mg/kg) and/or CCl4 (1 ml/kg 1:1 v/v CCl4: olive oil) in Control group, + groups 2 times per week for 4 consecutive weeks. caused significantly (p < 0.05) elevated serum activities alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine (ALT), total bilirubin (TB), whereas the albumin (ALB) protein (TP) levels reduced relative to control group. The catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) decreased 0.05), while malondialdehyde (MDA) level evidently comparison exposure significant increases proinflammatory interleukin-6 (IL-6) tumor necrosis factor-alpha (TNF-α), apoptotic caspase-3 caspase-9 levels, anti-inflammatory interleukin-4 (IL-4) interleukin-10 (IL-10) consistent histopathological changes compared control. On contrary, administration prevented toxicity lesions restoration status markers, antioxidant enzyme activities, MDA, cytokines apoptosis Altogether, findings reveal abrogate CCl4-provoked via inhibition stress, inflammation rats.

In Vivo and Computational Studies on Sitagliptin’s Neuroprotective Role in Type 2 Diabetes Mellitus: Implications for Alzheimer’s Disease DOI Creative Commons

Background/Objectives: Diabetes mellitus (DM), a widespread endocrine disorder characterized by chronic hyperglycemia, can cause nerve damage and increase the risk of neurodegenerative diseases such as Alzheimer’s disease (AD). Effective blood glucose management is essential, sitagliptin (SITG), dipeptidyl peptidase-4 (DPP-4) inhibitor, may offer neuroprotective benefits in type 2 diabetes (T2DM). Methods: T2DM was induced rats using nicotinamide (NICO) streptozotocin (STZ), biomarkers AD DM-linked enzymes, inflammation, oxidative stress, apoptosis were evaluated brain. Computational studies supported vivo findings. Results: SITG significantly reduced brain enzyme levels acetylcholinesterase (AChE), beta-secretase-1 (BACE-1), DPP-4, glycogen synthase kinase-3β (GSK-3β) T2DM-induced rats. It also inflammation lowering cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), tumor necrosis factor-α (TNF-α), nuclear factor-κB (NF-κB). Additionally, improved stress markers reducing malondialdehyde (MDA) enhancing glutathione (GSH). increased anti-apoptotic B-cell lymphoma protein-2 (Bcl-2) while pro-apoptotic Bcl-2-associated X (BAX) Caspace-3. lowered plasma insulin levels. To explore potential molecular level mechanisms, docking performed on AChE, COX-2, GSK-3β, BACE-1, The binding affinity for above-mentioned target enzymes 10.8, 8.0, 9.7, 7.7, 7.9 kcal/mol, respectively, comparable to co-crystallized ligands. Further mode analysis lowest energy conformation revealed interactions with critical residues. Conclusions: These findings highlight SITG’s targets T2DM-associated neurodegeneration its therapeutic approach AD, warranting further clinical investigations.

The possible protective effect of sitagliptin (SIT) against nephrotoxicity induced by a single dose 5-fluorouracil (5-FU) (150 mg/kg, i.p.) was investigated in rats. SIT treatment (5 and 10 mg/kg/day, p.o.) given for 7 days, starting 5 days before 5-FU administration.
