Due to the size and opacity of vertebrate brains, it has until now been impossible simultaneously record neuronal activity at cellular resolution across entire adult brain. As a result, scientists are forced choose between cellular-resolution microscopy over limited fields-of-view or whole-brain imaging coarse-grained resolution. ABSTRACT Animals integrate information from different sensory modalities as they mature and perform increasingly complex behaviors. This may parallel differential investment in specific brain regions depending on the changing demands of inputs. Abstract In vivo imaging of large-scale neuronal activity plays a pivotal role in unraveling the function brain's circuitry. Multiphoton microscopy, powerful tool for deep-tissue imaging, has received sustained interest advancing its speed, field view and depth. However, to avoid thermal damage scattering biological tissue, decreases exponentially as depth increases. We present suite innovations optimize three-photon microscopy large field-of-view at depths unreachable by two-photon microscopy. These techniques enable us image activities transgenic animals expressing protein calcium sensors ~ 3.5-mm diameter with single-cell resolution deepest cortical layer mouse brains. further demonstrate simultaneous subcortical brain, whole-brain adult zebrafish. The demonstrated can be integrated into typical multiphoton microscopes enlarge system-level neural circuit research.

Danionella cerebrum is a new vertebrate model that offers an exciting opportunity to visualize dynamic biological processes in intact adult animals. Key advantages of this include its small size, life-long optical transparency, genetic amenability and short generation time. Establishing reliable method for longitudinal vivo imaging D. while maintaining viability will allow in-depth image-based studies various involved development, disease onset progression, wound healing, aging live animal.
