Mammalian microbiomes have coevolved with their host to establish a stable homeostatic relationship. Multifaceted commensal-host and commensal–commensal interactions contribute the maintenance of equilibrium an impact on diverse physiological processes. Despite constant exposure physical chemical insults from environment, skin harbors surprisingly microbiome. The fungal compartment microbiome, mycobiome, is unique in that it dominated by single fungus, Malassezia. lack diversity suggests may provide niche for this genus Malassezia efficiently outcompete other fungi skin. This opinion article examines aspects support hypothesis, discusses how changes conditions associate mycobiome dysregulation, highlights emerging example being displaced pathogen C. auris, thereby generating predisposing situation fatal-invasive infection.

Immune checkpoint inhibitors (ICIs) activate anti-cancer immunity by blocking T cell molecules such as programmed death 1 (PD-1) and cytotoxic lymphocyte-associated protein 4 (CTLA-4). Although ICIs induce some durable responses in various cancer patients, they also have disadvantages, including low response rates, the potential for severe side effects, high treatment costs. Therefore, selection of patients who can benefit from ICI is critical, identification biomarkers essential to improve efficiency ICIs. In this review, we provide updated information on established predictive (tumor death-ligand [PD-L1] expression, DNA mismatch repair deficiency, microsatellite instability high, tumor mutational burden) currently under investigation tumor-infiltrated peripheral lymphocytes, gut microbiome, signaling pathways related damage antigen presentation. particular, review aims summarize current knowledge biomarkers, discuss issues, further explore future biomarkers.

The microbiome, a diverse ecosystem of microorganisms within the human body, interacts intricately with immune system to maintain health and prevent disease. This dynamic partnership influences development, inflammation regulation, metabolic health, neurological well‐being, while disruptions, termed dysbiosis, have been linked autoimmune disorders, chronic diseases, mental issues. review highlights microbiome’s role in educating system, modulating responses, supporting physical barriers, addressing key research advances, such as microbial metabolites regulation impact on cancer immunotherapy. Emerging technologies like multiomics machine learning propelled microbiome research, unlocking potential for personalized medicine microbiome‐targeted therapies. Despite progress, challenges remain understanding diversity, individual variability, causation–correlation microbiome–immune interactions. Future directions include precision medicine, engineering, public interventions, interdisciplinary collaborations, positioning symbiosis transformative frontier healthcare innovation.

Abstract Aging is associated with low‐grade inflammation that increases the risk of infection and disease, yet underlying mechanisms remain unclear. Gut microbiota composition shifts age, harboring microbes varied immunogenic capacities. We hypothesized gut acts as an active driver during aging. Microbiome patterns in aged mice strongly signs bacterial‐induced barrier disruption immune infiltration, including marked increased levels circulating lipopolysaccharide (LPS)‐binding protein (LBP) colonic calprotectin. Ex vivo immunogenicity assays revealed both contents mucosa harbored capacity to activate toll‐like receptor 4 (TLR4) whereas TLR5 signaling was unchanged. found elevated innate inflammatory (colonic Il6 , Tnf Tlr4 ) endotoxemia (circulating LBP) young germ‐free after weeks colonization intestinal from compared counterparts, thus providing a direct link between aging‐induced host inflammation. Additionally, we discovered exhibited unique responses broad‐spectrum antibiotic challenge (Abx), sustained elevation Escherichia (Proteobacteria) altered 7 days post‐Abx cessation. Together, these data indicate old age results differentially on TLR pathways system. age‐associated signatures are less resilient linked status. should be considered critical factors mediating chronic diseases disproportionally impacting older populations.
