Physical exercise effectively prevents anxiety disorders caused by environmental stress. The neural circuitry mechanism, however, remains incomplete. Here, we identified a previously unrecognized pathway originating from the primary motor cortex (M1) to medial prefrontal (mPFC) via ventromedial thalamic (VM) nuclei in male mice. Besides anatomical evidence, both ex vivo and recordings showed enhanced excitability of M1-VM inputs prelimbic (PrL) region mPFC upon 14-day treadmill on chronic restraint stress (CRS) mouse model. Further functional interrogations demonstrated that activation this circuit is necessary sufficient direct anxiolytic effect training CRS Our findings provide more insights into circuits connecting mental regions under paradigm implicate potential targets for neuromodulation treating disorders. relieves disorders, while authors show input cortex, nuclei.

Introduction In response to the global impact of COVID-19 pandemic, concerns about mental health, particularly anxiety levels, have become prominent. This study aims explore relationship between neuroticism, a personality trait associated with emotional instability, and during outbreak. Methods A comprehensive literature search was conducted using Cochrane Library, HINARI, Google Scholar, PUBMED, resulting in identification 26 relevant papers. The protocol has been registered PROSPERO under number CRD42023452418. Thorough meta-analysis performed Comprehensive Meta-Analysis V4 software. Results Meta-analysis revealed significant positive studies supporting this association (OR = 3.213, 95% CI 2.352 4.391). findings underscore importance considering traits, understanding psychological responses major crises such as epidemic. Discussion observed connection neuroticism heightened levels emphasizes need for targeted interventions, especially individuals high neuroticism. Further research into potential therapeutic approaches mitigating consequences context catastrophe is warranted. Systematic Review Registration https://www.crd.york.ac.uk/prospero/#CRD42023452418 .

Abstract Since the onset of COVID-19 pandemic, there have been concerns over mental health impact COVID-19. This is a review utilization antidepressants, anxiolytics, and hypnotics since pandemic was declared on March 11th 2020. A number reports so far based large prescription databases for administrative use at national or regional level, but mainly in high-income countries. We found studies reporting increased rates during 2020, which has interpreted as hoarding such medications. In following months, most antidepressant did not display clear pattern change compared with prepandemic trends. later phases small increases higher than predicted rates, consistent finding, especially youth. countries, were increasing trends antidepressants also before needs to be considered when estimating whereas anxiolytics hypnotics, patterns prescriptions more varying. Overall, after 2020 we could find any distinct changes pandemic. Most contain information about prevalence indicated psychiatric disorders studied populations. More are needed long-term effects COVID-19, particularly regarding antidepressants. Research relating major depression anxiety would promote better understanding how well reflect population.

Depression is one of the most serious chronic psychiatric disorders affecting people worldwide. Subthreshold depression (SD) a form subclinical with increased risk major depressive disorder (MDD). Patients diagnosed SD may not be eligible for antidepressant drugs and, particularly in case MDD, these antidepressants have adverse effects which outweigh their therapeutic effects, leading to discontinuation therapy. Food supplements could provide an alternative strategy. The aim this study demonstrate efficacy food supplement based on combination S-adenosyl methionine (SAMe, 200 mg/day) and probiotics (Lactobacillus helveticus Rosell®-52, Bifidobacterium longum Rosell®-175, 3 ×109 CFU/day) reducing symptoms monocentric, randomised, double-blind, placebo-controlled, cross-over clinical trial. 80 Subjects were recruited offered or placebo daily three months, according trial design, followed by six-week follow-up period. was measured means "Hamilton Rating Scale" (HAM-D) "Patient Health Questionnaire-9" (PHQ-9), using mixed analysis variance model, random intercept, statistical analysis. showed significant decrease PHQ-9 HAM-D scores resulting reduced MDD as compared placebo. In conclusion, intake SAMe probiotic strains period months effective improving quality life subjects who are therapies, patients suffering from mild-to-moderate sensitive cannot tolerate conventional drugs.

Abstract The major anxiety disorders (ANX; including generalized disorder, panic and phobias ) are highly prevalent, often onset early, persist throughout life, cause substantial global disability. Although distinct in their clinical presentations, they likely represent differential expressions of a dysregulated threat-response system. Here we present genome-wide association meta-analysis comprising 122,341 European ancestry ANX cases 729,881 controls. We identified 58 independent significant risk variants 66 genes with robust biological support. In an sample 1,175,012 self-report 1,956,379 controls, 51 the associated were replicated. As predicted by twin studies, found genetic correlation between depression, neuroticism, other internalizing phenotypes. Follow-up analyses demonstrated enrichment all brain regions highlighted GABAergic signaling as one potential mechanism underlying risk. These results advance our understanding architecture prioritize for functional follow-up studies.

Background Ecological momentary assessment (EMA) is a measurement methodology that involves the repeated collection of real-time data on participants’ behavior and experience in their natural environment. While EMA allows researchers to gain valuable insights into dynamic behavioral processes, need for frequent self-reporting can be burdensome disruptive. Compliance with protocols important accurate, unbiased sampling; yet, there no “gold standard” study design promote compliance. Objective The purpose this was use factorial identify optimal factors, or combinations achieving highest completion rates smartphone-based EMAs. Methods Participants recruited from across United States were randomized 1 2 levels each 5 factors 2×2×2×2×2 (32 conditions): factor 1—number questions per survey (15 vs 25); 2—number EMAs day (2 4); 3—EMA prompting schedule (random fixed times); 4—payment type (US $1 paid payment based percentage completed); 5—EMA response scale (ie, slider-type Likert-type scale; only within-person factor; participant complete slider- first 14 days second period).
