Abstract Transdermal delivery of drugs offers an interesting alternative for the administration molecules that present certain troubles when delivered by oral route. It can produce systemic effects or perform a local action formulation exerts optimal controlled drug release targeted to specific cell type site. also avoids several inconveniences such as hepatic first pass effect, gastric pH‐induced hydrolysis, malabsorption because diseases surgeries, and unpleasant organoleptic properties. Nanomedicine microneedle array patches (MAPs) are two trendiest systems applied transdermal research nowadays. However, skin is protective barrier nanoparticles (NPs) cannot through intact stratum corneum . The association NPs MAPs (NPs@MAPs) work synergistically, since assist bypass outer layers, contribute system providing delivery. Vaccination tailored therapies have been proposed fields where both great potential due inherent characteristics. conception easy use could allow self‐administration therefore facilitate mass vaccination campaigns in undeveloped areas with weak healthcare services. A new method is developed using elastic lipid nanovesicles (ELNs) loaded with ethanolic extract of Lantana camara (LC) to enhance skin permeation plant actives. The ELNs contained cholesterol, 1, 2-distearoyl-sn-glycero-3-phosphocholine, span 80, and tween 80. Firstly, 15 formulations were produced examine critical factors likely affecting formulation characteristics. In addition, surface characteristics, vesicle size, polydispersity index, zeta potential, degree deformability, % entrapment efficiency (% EE) ELN examined. As a significant parameter, was measured (using Start-M; it highly resembles human skin). influence hydrophilic-lipophilic balance (HLB), ratio vital through Start-M. particular, the LC decreased at more extensive size ranges, 400, 350-450 nm. contrast, increased significantly smaller such as 200 100 More than 75% permeated within 8 h when surfactant (span 80:tween 80; 25%:75%). Permeation studies conducted based on HLB values revealed that 78% in 12.2, an increase HLB. Cell viability assay SK-MEL-37 cells (skin cancer) reduced by ~80% 24 h, further validating formulation. Future research could investigate long-term safety therapeutic potential these clinical settings their effectiveness delivering other plant-based extracts for transdermal applications via ELNs.

Abstract Several drugs can be used for treating inflammatory skin pathologies like dermatitis and psoriasis. However, the management of chronic long-term cases, topical administration is preferred over oral delivery since it prevents certain issues due to systemic side effects from occurring. Cyclosporin A (CsA) has been this purpose; however, its high molecular weight (1202 Da) restricts diffusion through structure. Here, we developed a nano-in-micro device combining lipid vesicles (LVs) dissolving microneedle array patches (DMAPs) targeted delivery. CsA-LVs allowed effective incorporation CsA in hydrophilic DMAP matrix despite hydrophobicity drug. Polymeric composed poly (vinyl alcohol) (5% w/v), pyrrolidine) (15% w/v) CsA-LV dispersion (10% v/v) led formation CsA-LVs@DMAPs with adequate mechanical properties penetrate stratum corneum barrier.
