Glioblastoma (GBM) remains the epitome of aggressiveness and lethality in spectrum brain tumors, primarily due to blood–brain barrier (BBB) that hinders effective treatment delivery, tumor heterogeneity, presence treatment-resistant stem cells contribute recurrence. Nanoparticles (NPs) have been used overcome these obstacles by attaching targeting ligands enhance therapeutic efficacy. Among ligands, peptides stand out their ease synthesis high selectivity. This article aims review single multiligand strategies critically. In addition, it highlights other integrate effects external stimuli, biomimetic approaches, chemical approaches as nanocatalytic medicine, revealing significant potential treating GBM with peptide-functionalized NPs. Alternative routes parenteral administration, specifically nose-to-brain delivery local within resected cavity, are also discussed. Finally, an overview them discussed provide a perspective on this promising field therapy.

Nanocarrier drug delivery systems (NDDS) have gained momentum in the field of anticancer or nucleic acid due to their capacity aggrandize targeting efficacy and therapeutic outcomes encapsulated drugs. A disadvantage NDDS is that repeated administrations often encounter an obstacle known as "accelerated blood clearance (ABC) phenomenon". This phenomenon results rapid secondary dose from bloodstream markedly augmented liver accumulation, which substantially undermines accurate drugs effect NDDS. Nevertheless, underlying mechanism this has not been elucidated there currently no effective method for its eradication. In light above, aim review provide a comprehensive summary potential countermeasures ABC phenomenon, with view rejuvenating both slow-release property expectation clinic. paper, we innovatively introduce pharmacokinetic further elucidate occurrence after discussing immunological mechanism, provides new direction expanding mechanistic study phenomenon. Breast cancer is a fatal malignancy facing human health, with most patients experiencing recurrence and resistance to chemotherapy. The immunosuppressive tumor microenvironment (TME) greatly limits the actual outcome of immunotherapy. This study aimed develop modality theranostics nanoparticles for breast based on near-infrared light-triggered nanoparticle targeted delivery ginsenoside Rg3 immune adjuvants imiquimod (R837) effective Folate-receptor (FA) targeting IR780-R837/ginsenoside Rg3-perfluorohexane (PFH) @ polyethylene glycol (PEG)-poly (lactide-co-glycolic acid) (PLGA) (FA-NPs) can be activated by laser irradiation in tumors, which leads rapid release R837 regions high expression folate receptors glucose transporter 1 (GLUT1). Meanwhile, used as dual-mode contrast agents photoacoustic ultrasound imaging. strategy provides strong memory effect, prevent after eliminating initial tumor.

Abstract Background Ginsenoside Rg3 is a component of ginseng that protects against myocardial ischemia/reperfusion (MI/R) injury. Ferroptosis new form cell death characterized by oxidative damage to phospholipids. The purpose this study was examine the role and ginsenoside in MI/R mechanism. Methods A mouse model left anterior descending (LAD) ligation-induced injury oxygen-glucose deprivation/reperfusion (OGD/R) were used as vitro vivo models, respectively. Echocardiographic analysis, 2,3,5-triphenyltetrazolium chloride (TTC) staining hematoxylin-eosin (H&E) assess cardioprotective effects Rg3. Western blotting, biochemical small interfering RNA analysis molecular docking performed underlying Results improved cardiac function infarct size mice with Moreover, increased expression ferroptosis-related protein GPX4 inhibited iron deposition also activated Nrf2 signaling pathway. attenuated ischemia/reperfusion-induced ferroptosis via Notably, regulated keap1/Nrf2 pathway attenuate OGD/R-induced H9C2 cells. Taken together, keap1/Nrf2/GPX4 Conclusions Our findings demonstrated ameliorate MI/R-induced

Polyion complex (PIC) nanoparticles, including PIC micelles and PICsomes, are typically composed of poly(ethylene glycol) block copolymers coupled with oppositely charged polyelectrolytes or therapeutic agents via electrostatic interaction. Due to a simple rapid preparation process high drug-loading efficiency, nanoparticles beneficial maintaining the chemical integrity biological activity loaded drugs. However, stability can be disrupted in high-ionic-strength solutions because interaction is DRIVING force; these disruptions thus impair drug delivery. Herein, we summarize advances use for delivery drugs, focusing on different physical strategies employed enhance their stability, enhancing charge density, crosslinking, increasing hydrophobic interactions, forming hydrogen bonds, development PIC-based gels. In particular, describe load peptide antibiotics targeting antibiotic-resistant biofilm-related diseases that chemotherapeutics gaseous donors cancer treatment. Furthermore, application as magnetic resonance imaging contrast summarized first time. Therefore, this review great significance polymeric functional
