Fuzi is a famous toxic traditional herbal medicine, which has long been used for the treatment of various diseases in China and many other Asian countries because its extraordinary pharmacological activities high toxicity. Diabetes cardiomyopathy (DCM) is a metabolic disease and leading cause of heart failure among people with diabetes. Mass spectrometry imaging (MSI) versatile technique capable combining the molecular specificity mass spatial information imaging. In this study, we used MSI to visualize metabolites in rat high resolution sensitivity. We optimized air flow-assisted desorption electrospray ionization (AFADESI)-MSI platform detect wide range metabolites, then matrix-assisted laser (MALDI)-MSI for increasing coverage improving localization resolution. AFADESI-MSI detected 214 149 positive negative analyses sections, respectively, while MALDI-MSI 61 analysis. Ziziphi Spinosae Semen (ZSS), a medicinal food, is one of the most popular Chinese herbs, known for its rich bioactive ingredients, such as terpenoids and flavonoids. The content these ingredients in ZSS varies with age. However, distribution throughout developmental phases ZSS-white maturity (S1), firm ripening (S2), full (S3)-and within specific tissues like cotyledon, endosperm, radicle, has not yet been determined. This research aimed to analyze spatial key quality markers elucidate metabolite characteristics growth stages. Utilizing matrix-assisted laser desorption/ionization-mass spectrometry imaging (MALDI-MSI), 25 were identified visualized ZSS, including three terpenoids, seven flavonoids, four alkaloids, eleven amino acids.
