The inflammatory response consists of two stages: priming and triggering. triggering stage is marked by the activation inflammasomes, which are cytosolic protein complexes acting as platforms for inflammation. Inflammasomes divided into canonical noncanonical categories. Inflammatory lung diseases such asthma, chronic obstructive pulmonary disease (COPD), acute respiratory distress syndrome (ARDS), injury, fibrosis arise from inflammation damage. While role inflammasomes in these well demonstrated, recent findings emphasize critical roles regulating various conditions. Particularly, new studies highlight their involvement diseases. This review delves research on regulatory human caspase-4 murine caspase-11, development diseases, potential targeting treatments.

Postoperative cognitive dysfunction (POCD) is a cluster of problems that may arise after surgery. POCD symptoms include memory loss, focus inattention, and communication difficulties. Inflammasomes, intracellular multiprotein complexes control inflammation, have significant role in the development POCD. It has been postulated NLRP3 inflammasome promotes impairment by triggering inflammatory response brain. Nevertheless, there are many gaps current literature to understand underlying pathophysiological mechanisms develop future therapy. The Artemisia argyi Lévl. et Van. growing in the surrounding areas of Qichun County China are called Qiai (QA). is a crop that can be used both as food and traditional folk medicine. Quercetin is a natural polyphenolic flavonoid widely found in plants, fruits, and vegetables, has been reported to play pharmacological roles numerous pathogenic conditions. The anti-inflammatory effects of quercetin various inflammatory conditions diseases have well-documented. However, its regulatory role noncanonical inflammasome activation not yet demonstrated. This study investigated the caspase-11 inflammasome-activated responses macrophages mouse model acute lethal sepsis. protected J774A.1 from lipopolysaccharide (LPS)-induced cell death inflammasome-induced pyroptosis. It significantly decreased production mRNA expression pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-18, IL-6, but tumor necrosis factor (TNF)-α, molecules, nitric oxide (NO) inducible NO synthase cells. Mechanistically, strongly suppressed autoproteolysis secretion proteolysis gasdermin D did inhibit direct binding LPS. In vivo, revealed that increased survival rate mice with sepsis serum levels cytokines without causing significant toxicity. conclusion, this highlights quercetin-mediated action through novel mechanism targets macrophages, suggesting promising agent medicine.

Among the plants that exhibit significant or established pharmacological activity, genus Artemisia L. deserves special attention. This comprises over 500 species belonging to largest Asteraceae family. Our study aimed at providing a comprehensive evaluation of phytochemical composition ethanol extracts five different (collected from southwest Russian Federation) and their antimicrobial nematocide activity as follows: A. annua cv. Novichok., dracunculus Smaragd, santonica Citral, abrotanum Euxin, scoparia Tavrida. The using methods gas chromatography–mass spectrometry (GC–MS) high-performance liquid chromatography–quadrupole time-of-flight mass (HPLC–MS/MS) allowed establishing profile. obtained data on extracts’ were used predict antibacterial antifungal against phytopathogenic microorganisms nematocidal free-living soil nematode Caenorhabditis elegans. major compounds found in monoterpenes, sesquiterpenes, flavonoids, flavonoid glycosides, coumarins, phenolic acids. began manifest concentration 150 µg/mL. Smaragd extract had selective effect Gram-positive R. iranicus B. subtilis bacteria, whereas Tavrida Gram-negative tumefaciens X. arboricola bacteria solani, solani F. graminearum fungi. Novichok, Citral range 31.3–1000 µg/mL caused death nematodes. It was Novichok affects UNC-63 protein, molecular target which is nicotine receptor N-subtype.

Cecropin AD (CAD), a renowned antimicrobial peptide, has shown promising potential in treating various bacterial infections. This study investigates the protective effects of CAD against lipopolysaccharide (LPS)-induced intestinal adversities chickens. Sixty SPF-grade chicks were divided into groups and exposed to different dosages CAD, followed by LPS administration. The assessed impact on mucosal injury markers, oxidative stress, inflammation. significantly increased Diamine oxidase (DAO) D-lactate (D-LA) levels, both indicators injury. treatment substantially attenuated these elevations, particularly at higher dosages. Additionally, markedly reduced stress tissues, as normalized antioxidant levels decreased reactive oxygen species. Histological analysis supported findings, showing better-preserved villi structures CAD-treated groups. Furthermore, IL-6 IL-8 expression post-LPS stimulation effectively regulated NLRP3 inflammasome pathway, decreasing associated factors like NLRP3, Caspase-1, IL-1b, IL-18. demonstrates CAD's therapeutic alleviating LPS-induced injuries. are primarily attributed its anti-inflammatory antioxidative actions modulation pathway.
