Ricin is a highly toxic plant toxin that can cause multi-organ failure, especially liver dysfunction, and potential bioterrorism agent. Despite the serious public health challenge posed by ricin, effective therapeutic for ricin-induced poisoning currently unavailable. Nickel oxide nanoparticles (NiONPs) are toxic heavy metal compounds that induce liver fibrosis and metabolic disorders. Current research shows the intestinal microbiota regulates metabolism through gut-liver axis. As one of the neonicotinoid insecticides, thiacloprid (THI) is extensively used in agriculture and frequently detected various aquatic environments, posing a potential threat to organisms. However, effects THI exposure on turtles remain unknown. In this study, we focused investigating whether has toxic effect gut-liver axis turtles. The Reeves' (Mauremys reevesii) were exposed 0.0178 μM, 6 60 μM for 5 consecutive weeks. results revealed that altered composition intestinal flora, with decrease relative abundance Romboutsia, an increase Clostridium_sensu_stricto_1, Cetobacterium, Enterococcus. Ambient ozone (O3) pollution can induce respiratory and cardiovascular toxicity. However, its impact on the metabolome underlying mechanisms remain unclear. This study first investigated serum metabolite changes in rats exposed to 0.5 ppm O3 for 3 months using untargeted metabolomic approach. Results showed chronic exposure significantly altered levels of 34 metabolites with potential increased risk digestive, disease. Moreover, bile acid synthesis secretion, arachidonic (AA) metabolism became most prominent affected metabolic pathways after exposure. Further studies found that elevated toxic was not due biosynthesis liver, but reduced reuptake from portal vein hepatocytes owing repressed Ntcp Oatp1a1, decreased efflux as a results inhibited Bsep, Ostalpha Ostbeta. Meanwhile, expressions detoxification enzyme SULT2A1 important regulators FXR, PXR HNF4α also contributed abnormal acids. In addition, promoted conversion AA into thromboxane A2 (TXA2) 20-hydroxyarachidonic (20-HETE) liver by up-regulation Fads2, Cyp4a Tbxas1 which resulting linoleic (LA), B2 (TXB2) 20-HETE serum. Excess acetaminophen can result in hepatotoxicity, while luteolin has the anti-liver injury property. However, low water solubility reduces its pharmacological performance. Luteolin-β-Cyclodextrin-Metal-Organic-Framework (Luteolin-β-CD-MOF) is an alternative with more water-soluble activity of luteolin. In this study, we utilized vivo and vitro methods to explore underlying protective mechanism luteolin-β-CD-MOF acetaminophen-induced liver injury. Untargeted metabolomics were performed, along study bile acids metabolism injured rats. Western blotting assays performed assess expressions ferroptosis-related proteins (glutathione peroxidase 4 (GPX4), ferritin heavy chain-1 (FTH1), heme oxygenase-1 (HO-1), solute carrier family 7 member 11 (SLC7A11)) rats L02 cells. ABSTRACT The mechanism of anthocyanin in alleviating intestine injury pigs is worth pursuing due to the higher similarity physiological structure between human beings and than rodents. Herein, we investigated by which improves gut damage induced with lipopolysaccharide further confirmed this using fecal microbiota transplantation. Thirty‐two piglets were randomly divided into four treatments: control group (Control), model (LPS), prophylactic (An‐LPS), transplantation (FMT‐LPS). results showed that oxidative stress increased expression p‐p65, Bax, Keap1 decreased Nrf2, Occludin, Claudin‐1 jejunum. However, its suspension reversed these influences. Meanwhile, selectively improved certain microbiota, including Lactococcus , leading distinct metabolic profiles. These differences favor activating antioxidant‐related signaling pathway, Nrf2/Keap1 inhibiting pro‐inflammatory pathway such as NF‐κB, alleviated inflammation significantly intestinal tight junction, Occludin Claudin‐1. Moreover, compared LPS, serum antioxidant enzyme activity IL‐10 content An‐LPS. In conclusion, ameliorated through antioxidant‐ inflammation‐related pathways; meanwhile, metabolites play a nonsubstitutable role process.
