Circadian clocks may mediate lifespan extension by caloric or dietary restriction (DR). We find that the core clock transcription factor Clock is crucial for a robust longevity and fecundity response to DR in Drosophila . To identify clock-controlled mediators, we performed RNA-sequencing from abdominal fat bodies across 24 h day after just 5 days under control diets. In contrast more chronic regimens, did not detect significant changes rhythmic expression of genes. Yet discovered induced de novo rhythmicity increased output Network analysis revealed network connectivity one module comprised genes encoding proteasome subunits. Adult, body specific RNAi knockdown demonstrated subunits contribute DR-mediated extension. Thus, links

Vascular dementia (VaD) is the second most common type of dementia. VaD synonymous with ageing, and its symptoms place a significant burden on health wellbeing older people. Despite identification substantial number risk factors for VaD, pathological mechanisms underpinning this disease remain to be fully elucidated. Consequently, biogerontological imperative exists highlight modifiable lifestyle which can mitigate against developing VaD. This review will critically examine some have been revealed modulate risk. The survey commences by providing an overview putative are associated pathobiology Next, influence examined. Finally, emerging treatment avenues including epigenetics, gut microbiome, pro-longevity pharmaceuticals discussed. By drawing key evidence together, it our hope that used inform future experimental investigations in field.

Ageing constitutes the biggest risk factor for poor health and adversely affects integrity function of all cells, tissues, organs in human body. Vascular ageing, characterised by vascular stiffness, endothelial dysfunction, increased oxidative stress, chronic low-grade inflammation, early-stage atherosclerosis, may trigger or exacerbate development age-related diseases, which each year contribute to more than 3.8 million deaths Europe alone necessitate a better understanding mechanisms involved. To this end, large number recent preclinical clinical studies have focused on exponential accumulation senescent cells system paid particular attention specific roles senescence-associated secretory phenotype, proteostasis age-mediated modulation certain microRNA (miRNAs), contribution other major factors, notably diabetes, hypertension, smoking, ageing elderly. The data generated paved way various senotherapeutic interventions, ranging from application synthetic natural senolytics senomorphics attempt modify lifestyle, control diet, restrict calorie intake. However, guidelines, considering severity characteristics need be established before widespread use these agents. This review briefly discusses molecular cellular summarises efficacy widely studied senotherapeutics context ageing.

Limited resources affect an organism's physiology through the conserved metabolic pathway, mechanistic target of rapamycin (mTOR). Males and females often react differently to nutritional limitation, but whether it leads differential mTOR pathway expression remains unknown. Recently, we found that dietary restriction (DR) induced significant changes in genes female Japanese quails (Coturnix japonica). We simultaneously exposed 32 male either 20%, 30%, 40% or ad libitum feeding for 14 days determined six key liver investigate sex differences patterns. DR significantly reduced body mass, albeit effect was milder males compared females. observed sex-specific gene expression. downregulated more than males. Under moderate DR, ATG9A RPS6K1 expressions were increased Like females, mass correlated positively with IGF1, negatively RS6K1 expressions. Our findings highlight sexes may cope deficits emphasise importance considering sexual studies restriction.

ABSTRACT The ability of organisms to cope with poor quality nutrition is essential for their persistence. For species a short generation time, the nutritional environments can transcend generations, making it beneficial adults prime offspring particular diets. However, our understanding adaptive generational responses, including those diet quality, are still very limited. Here, we used vinegar fly, Drosophila melanogaster, investigate whether females developing as larvae on nutritionally produce that primed nutrient deficiencies in following generations. We found developed low-quality diets produced that, similarly diets, had both increased egg-to-adult viability and starvation tolerance compared experiencing nutrient-rich diet. When testing persistence such priming, just one high-quality sufficient erase signal priming. A global transcriptomic profile analysis male suggests observed phenotypic priming not constitutive adjustment adults; instead, probably larvae, enabling them initiate an response when exposed Our results support important mechanism enables transient fluctuations.

This study investigates the association between dietary methyl donor nutrients intake and phenotypic aging. cross-sectional comprised 27,211 adult participants from NHANES 2005-2018. The methyl-donor nutritional quality index (MNQI) was calculated by assessing of seven nutrients: protein, folate, choline, riboflavin, vitamin B6, B12, zinc. Phenotypic age acceleration (PhenoAge.Accel) using biochemical markers to assess biological Weighted generalized linear regression models were utilized associations MNQI PhenoAge.Accel, impact different demographic health characteristics evaluated through interaction effect tests. After adjusting for various potential confounding factors, a significant negative found PhenoAge.Accel (β = - 0.66; 95% CI 0.91, 0.40; P < 0.0001), indicating that an increase in is associated with slowdown PhenoAge.Accel. Furthermore, subgroup analysis indicated stronger negatively males 0.94; 1.36, 0.52) interactions (Pinteraction 0.02). In populations, PhenoAge Accel, suggesting choosing high-quality foods can help develop effective strategies enhance healthy longevity.

In the last decade epigenetics has come to fore as a discipline which is central biogerontology. Age associated epigenetic changes are routinely linked with pathologies, including cardiovascular disease, cancer, and Alzheimer's disease; moreover, clocks capable of correlating biological age chronological in many species humans. Recent intriguing empirical observations also suggest that inherited effects could influence lifespan/longevity variety organisms. If this case, an imperative exists reconcile processes evolution ageing. This review will critically evaluate from evolutionary perspective.
