Neuron activity across the brain How is it that groups of neurons dispersed through interact to generate complex behaviors? Three papers in this issue present brain-scale studies neuronal and dynamics (see Perspective by Huk Hart). Allen et al. found thirsty mice, there widespread neural related stimuli elicit licking drinking. Individual encoded task-specific responses, but every area contained with different types response. Optogenetic stimulation thirst-sensing one reinstated drinking previously signaled thirst. Gründemann investigated mouse basal amygdala relation behavior during tasks. Two ensembles showed orthogonal exploratory nonexploratory behaviors, possibly reflecting levels anxiety experienced these areas. Abstract Single-cell RNA sequencing (scRNA-seq) provides details for individual cells; however, crucial spatial information is often lost. We present SpaOTsc, a method relying on structured optimal transport to recover properties of scRNA-seq data by utilizing measurements relatively small number genes. A metric cells in first established based map connecting it with the measurements. The cell–cell communications are then obtained “optimally transporting” signal senders target receivers space. Using partial decomposition, we next compute intercellular gene–gene flow estimate regulations between genes across cells. Four datasets employed cross-validation gene expression prediction and comparison known communications. SpaOTsc has broader applications, both integrating non-spatial single-cell data, directly transcriptomics reconstruct cellular dynamics tissues.

We present a unique, extensive, and open synaptic physiology analysis platform dataset. Through its application, we reveal principles that relate cell type to properties intralaminar circuit organization in the mouse human cortex. The dynamics of excitatory synapses align with postsynaptic subclass, whereas inhibitory synapse partly presynaptic subclass but considerable overlap. Synaptic are heterogeneous most subclass-to-subclass connections. two main axes heterogeneity strength variability. Cell subclasses divide along variability axis, axis accounts for substantial within subclass. In cortex, excitatory-to-excitatory distinct from those cortex vary depth across layers 2 3.

Neural activity and behavior are both notoriously variable, with responses differing widely between repeated presentation of identical stimuli or trials. Recent results in humans animals reveal that these variations not random their nature, but may fact be due large part to rapid shifts neural, cognitive, behavioral states. Here we review recent advances the understanding waking state, how generated, they modulate neural mice humans. We propose brain has an identifiable set states through which it wanders continuously a nonrandom fashion, owing ascending modulatory fast-acting corticocortical subcortical-cortical pathways.
